HPLC-ELSD法测定溃疡灵胶囊中黄芪甲苷的含量

摘　　要：目的建立溃疡灵胶囊中黄芪甲苷的含量测定方法。方法采用HPLC-ELSD法对溃疡灵胶囊中黄芪甲苷进行定量分析,色谱柱:Wa-tersC18(4.6mm×150mm,5μm)。流动相:乙腈-水(35:65);流速:0.8mL.min-1;柱温:30℃;ELSD漂移管温度:85℃,载气为高纯氮气(99.999%):1.5L.min-1;放大系数(Gain):4。结果黄芪甲苷在1.05μg～3.68μg范围内呈良好的线性关系,r=0.9998,平均加样回收率为95.9%,RSD=1.6%。结论本法对黄芪甲苷的含量测定良好,可用于溃疡灵胶囊的质量控制。OBJECTIVE To establish a method for the determination of astragloside Ⅳ in Kuiyangling capsules.METHODS A new quantitative analysis of astragloside Ⅳ in Kuiyangling capsules was adopted by HPLC-ELSD.The column was Waters C18（4.6mm×150mm,5um）,with mobile phase of acetonitrile-water（35∶65）at the flow rate of 0.8 mL·min-1,the column temperature was 30℃.The drift tube temperature of the evaporative light-scattering detector was 85℃ and the nebulizer nitrogen flow rate was 1.5L·min-1,gain was 4.RESULTS The method had good linear relationship within the range of 1.05～3.68μg（r=0.9998）for astragloside Ⅳ,The average recoveries（n=9） were 95.9%（RSD=1.6）.CONCLUSION The method is well-done for the determination of astragloside Ⅳ.It can be used for quality control of Kuiyangling capsules.
