摘 要:目的观察过氧化物酶增殖活化受体γ(peroxisome proliferator-activated receptorγ,PPARγ)激动剂——吡格列酮对老龄鼠下颌牙槽骨骨代谢的影响。方法取18月龄健康雄性C57BL/6小鼠20只,随机分为两组,实验组每日予以吡格列酮20mg/kg灌胃,对照组每日予以等量生理盐水灌胃。8周后处死老龄鼠,取双侧下颌骨,行下颌骨X线检查及牙槽骨骨密度测定,牙槽骨组织形态学观察,采用骨形态计量学方法计算牙槽骨的静态骨参数。结果实验组牙槽骨X线片的阻射程度、骨密度、骨小梁面积百分率、骨小梁的平均宽度及总胶原的含量均明显低于对照组。结论外源性配体激活PPARγ能导致老龄鼠牙槽骨成骨及破骨代谢的失衡,促进老龄鼠牙槽骨骨质疏松的发生、发展。Objective To investigate the influence of peroxisome proliferator-activated receptor γ(PPARγ)on the alveolar bone of mandibles in aged mice by giving PPARγ agonist,Pioglitazone.Methods 20 mice aged 18 months were randomly divided into two groups.Mice in the experimental group received intragastric administration of 20 mg/kg Pioglitazone,and mice in the control group were treated with gastric perfusion of physiologic saline for 8 weeks.The effects of PPARγ were assessed by calculating the degree of the radiopacity after X-ray examination,examining the bone mineral density,the histological methods and measuring the shaped parameters of the alveolar bone.Results The degree of the radiopacity,the bone mineral density,the area of the alveolar bone and the total collagen of the experimental group were significantly lower than those of the control group.Conclusions Exogenous ligand of PPARγ can disrupt the balance between the bone formation and the bone absorption,promoting the progress of osteoporosis in alveolar bone in aged mice.
