摘  要:目的观察富血小板血浆(platelet-rich plasma,PRP)对体外培养的大鼠骨髓基质干细胞(bone marrow stromal cells,BMSCs)增殖和矿化的影响。方法采用贴壁法分离培养大鼠股骨来源的BMSCs,二次离心法提取大鼠富血小板血浆,采用含体积浓度为10%PRP的培养液培养BMSCs作为实验组,不含PRP者为对照组,检测大鼠BMSCs的增殖和矿化情况。结果 PRP组在MTT法检测中较对照组具有更高的吸光度(OD)值,但是其在碱性磷酸酶(alkaline phosphatase,ALP)活性值和矿化结节形成数上均低于对照组。结论富血小板血浆可促进大鼠BMSCs增殖,而早期可抑制其矿化。Objective To observe the effects of platelet-rich plasma(PRP)on the proliferation and mineralized accumμlation of bone marrow stromal cells(BMSCs)from rat cμltured in vitro.Methods BMSCs were isolated and proliferated by cμlture in vitro and the second passage BMSCs were divided into two groups,10% PRP group was added 10% PRP which was obtained by centrifuging for twice,the proliferation and mineralization of the two groups were compared.Results The mean absorbance value was significantly higher in the PRP group than in the control group at the MTT assay,however,compared with the control group,the PRP group demonstrated less alkaline phosphatase(ALP)activity and less mineralized nodμle formation.Conclusions PRP can promote the proliferation of rat's BMSCs but inhibit mineralization at early stage.
