L-NAME预处理对缺氧复合氰化钠中毒大鼠脑损伤影响

摘  要:目的观察非特异性NOS抑制剂L-NAME预处理后高原缺氧复合氰化钠中毒大鼠脑组织NOS、NO、SOD、MDA值改变,以SOD、MDA为指标评价脑缺氧损伤程度。方法 40只雄性SD大鼠随机划分为L-NAME预处理(3%L-NAME溶液30mg/kg,1次/d,皮下注射)及L-NAME未预处理组,再将每组随机均分为平原组、平原氰化钠中毒组、高原组、高原缺氧复合氰化钠中毒组。中毒组于皮下注射氰化钠3.6mg/kg,0.5h时相点处死动物,取脑检测NOS、NO、SOD、MDA值。结果 L-NAME预处理后,各组脑NOS活性、NO含量均显著性降低,高原缺氧复合氰化钠中毒组脑SOD活性显著性降低,MDA含量显著性增加。结论缺氧复合氰化钠中毒较单纯缺氧或中毒脑损伤加重。L-NAME预处理未明显改善单纯性缺氧或中毒组脑缺氧损伤,且有加重缺氧复合氰化钠中毒脑缺氧损伤作用。Objective To observe the changes of nitric oxide synthase ( NOS), NO, malondialdehyde (MDA) and superoxide dismutase(SOD) in cerebral tissues from a rat model of hypoxia combined with cyanide intoxication, and to evaluate the degree of brain hypoxic injury by using SOD and MDA as indicators. Methods A total of 40 male SD rats were randomly divided into L - NAME pretreatment group( subcutaneous injection of 3% L - NAME at 30 mg/kg once a day)and non - L - NAME pretreatment group,and then both groups were subdivided into plain group, plain group with NaCN intoxication, high altitude group( simulated 4000 m altitude) , and high altitude group with NaCN intoxication (simulated 4000 m altitude ). Rats in two NaCN intoxication subgroups were subcutaneously injected NaCN at 3.6 mg/kg while those without NaCN intoxication were injected normal saline. Half an hour later,all animals were killed and their brain tissues were removed for the determination of NOS activity, SOD activity, NO content and MDA content by spectrophotometric method. Results After L - NAME pretreatment, cerebral NOS activity and NO content of rats in each group decreased significantly, SOD activity in high altitude group with NaCN intoxication decreased significantly and MAD content in this group increased significantly. Conclusion The rat brain injuries caused by hypoxia combined with NaCN intoxication were more severe than those caused by simple hypoxia or NaCN intoxication. L - NAME pretreatment appears not to improve the brain injuries in rats suffering from simple hypoxia or intoxication, and it may aggravate the brain injuries in rats suffering acute hypoxia combined with NaCN intoxication.
