摘 要:以尾穗苋为试验材料,取其无菌苗子叶节为外植体,对苗龄为3d、5d、7d的不同子叶节苗态、切割方式以及激素配比等分化再生因素进行优化筛选。结果表明:不同子叶节苗态及切割方式对不定茅分化影响不大,其中子叶刚平展开的子叶节,采用子叶全留的切割方式有利于不定茅的生长;培养基激素配比对不定茅的诱导影响显著,单独使用6-BA能诱导子叶节产生不定芽,但芽生长较慢且弱小,NAA和6-BA配合使用能促进不定芽的生长和伸长,生长健壮,其中1.0mg/L 6-BA+0.2mg/L NAA组合对不定芽的诱导与生长效果最佳。MS培养基中添加0.2mg/L IBA诱导生根的效果最好,生根率可达97.8%。Cotyledonary nodes of A maranthus caudatus Linn. were used for tissue culture to establish a high-frequency regeneration system, and the effect of seedling stages, cutting ways and hormone combinations on differentiation of adventitious shoot and regeneration frequency were studied and optimized. The results showed that different seedling stages and cutting ways did not show distinct difference to the differentiation of shoots, and cotyledonarys that were flatly separated promoted the differentiation and growth of shoots. Single use of 6-BA could successfully induce shoots, but they grew slowly and weakly. While the combinations of 6-BA plus NAA were very effective in growing and elongating of shoots, and the best medium was MS+1.0mg/L 6-BA+0.2mg/L NAA. The IBA concentration at 0.2mg/L favored root formation, resulting in a rooting rate of up to 97.8%.
