促卵泡素对犬卵母细胞体外成熟的影响

摘　　要：为探究基础培养基中添加促卵泡素(FSH)对犬卵母细胞体外成熟的影响,采用切割法收集卵巢表面的卵丘卵母细胞复合体(COCs),培养在添加不同浓度FSH(0,0.1,1,10 IU/mL)的TCM-199基础培养液中.体外培养72 h后,对卵母细胞进行染色观察其细胞核成熟情况.结果表明,FSH的添加没有增加犬卵母细胞恢复减数分裂的细胞数量.此外,不同处理对卵母细胞卵丘扩散无差异.Canine oocytes underwent in vitro maturation（IVM） in culture medium containing Follicle Stimulation Hormone（FSH）was studied.Cumulus-oocytes complexes（COCs） were harvested from ovaries by slicing subjected to IVM in TCM-199 medium supplemented with varies concentrations of FSH（0,0.1,1,and10 IU/mL）.After culture for 72h,oocyte were stained and examined for nuclear maturation.Supplementation with FSH did not increase the number of oocytes that resumed meiosis.Additionally,there were no differences among treatments in the cumulus cell expansion of oocytes.
