摘  要:目的建立测定血浆中甲氨蝶呤浓度的反相高效液相色谱法,为临床合理用药提供参考。方法色谱柱采用Agilent TC-C18柱(250 mm×4.6 mm,5μm),流动相为0.025 mol/L磷酸二氢钠溶液(pH为5.41)-甲醇(76∶24,V/V),流速1.0 mL/min,检测波长313 nm,柱温35℃,样品经高氯酸沉淀蛋白后直接进样。结果血浆中甲氨蝶呤的质量浓度在0.05~5μg/mL和5~100μg/mL范围内与峰面积线性关系良好,r1=0.999 9,r2=0.999 8,平均回收率分别为98.06%和98.79%,最低定量质量浓度为0.05μg/mL,日内及日间精密度的RSD均小于4%(n=3)。结论所用方法简便快速、准确可靠,灵敏度高,重现性好,线性范围宽,成本低,适于甲氨蝶呤的血药浓度监测。Objective To set up a rapid RP-HPLC method to determine the plasma concentration of methotrexate(MTX).Methods The analytical column was Agilent TC-C18(250 mm × 4.6 mm,5 μm).The mobile phase was 0.025 mol /L NaH2PO4(pH = 5.41)-methanol(76 ∶ 24,V/V).The flow rate was 1.0 mL/min.The detection wavelength was 313 nm.The column temperature was set at 35 ℃.The plasma samples were deposited down the protein with 10% HClO4.Results The linear relation was in the range of 0.05-5 μg/mL(r1 = 0.999 9) and the range of 5-100 μg/mL(r2 = 0.999 8),respectively.The average recovery rates were 98.06% and 98.79%,respectively.The measurable lowest concentration was 0.05 μg/mL.RSD with intra-day and inter-day was less than 4%.Conclusion The established method is simple,accurate,sensitive and reproducible with large-scale of detection range,which is suitable for monitoring the blood concentration of methotrexate.
