阿托伐他汀对高胆固醇血症患者清道夫受体BⅠ表达的影响

摘  要:目的探讨清道夫受体BⅠ(SR-BⅠ)在动脉粥样硬化发生中的作用及阿托伐他汀抗动脉粥样硬化的作用机制。方法随机选择健康体检中心男性高TC血症患者60例为高脂组,另选男性TC正常者60例为对照组。采用紫外分光法检测血管紧张素转换酶(ACE)的活性,放射免疫法检测血管紧张素Ⅱ(AngⅡ)水平,硝酸还原酶法检测NO水平,RT-PCR和Western blot法分别检测SR-BⅠ mRNA和蛋白表达水平。结果与对照组比较,高脂组TC、TG、LDL-C、AngⅡ、ACE明显升高(P<0.05,P<0.01),HDL-C、NO、SR-BⅠ mRNA和蛋白表达明显降低(P<0.05,P<0.01);与同组治疗前比较,高脂组治疗后TC、TG、LDL-C、Ang Ⅱ、ACE明显降低(P<0.05),HDL-C、NO、SR-BⅠ mRNA和蛋白表达明显升高(P<0.05,P<0.01)。HDL-C与SR-BⅠ呈显著正相关,AngⅡ与SR-BⅠ呈显著负相关。结论高TC血症患者SR-BⅠ表达降低,阿托伐他汀能上调SR-BⅠ表达。Objective To investigate the role of scavenger receptor class B type I (SR-B I ) in genesis of atherosclerosis and the mechanism of antiatherosclerotic effects of statins by observing the changes of SR-B I expression and the effects of atorvastatin on them. Methods Sixty patients with hypercholesterolemia and sixty healthy persons were investigated as hyperlipidemia group (Hyperlipidemias) and control group(Controls) respectively. Hyperlipidemias were treated with atorvastatin 20 mg/d for twelve weeks. The level of angiotensin converting enzyme(ACE) in blood serum was determined by ultraviolet spectroscopy. The plasma levels of angiotensin II (Ang II ) and nitric oxide(NO) were detected by radioimmunoassay and nitrate reductase respectively. The expression of SR-B I mRNA and protein on platelet was assessed by RT-PCR and Western blot respectively. Results The serum ACE,TC,TG,LDL-C and plasma Ang II of Hyperlipidemias were higher than those of Controls (P 〈 0.01) ,but those of Hyperlipidemias were decreased after treatment. The levels of NO and HDL-C of Hyperlipidemias were obviously lower than those of Controls (P〈 0. 01), but increased after treatment. The expression of SR-B I mRNA and protein on platelets was lower in Hyperlipidemias than in Controls (P 〈0.05),but increased after treatment. The expression of SR-B I protein was positively correlated with the level of HDL-C and inversely correlated with the plasma AngII (P 〈 0.05) in patients with hy- percholesterolemia. Conclusions The expression of SR-B I decreases and atorvastatin upregulates its expression on platelets in patients with hypercholesterolemia.
