摘 要:目的建立高效液相色谱法(high performance liquid chromatography,HPLC)测定大鼠血浆中白藜芦醇浓度,并用该法研究大鼠静脉注射白藜芦醇后的药物代谢动力学。方法取大鼠血浆样品,以乙腈沉淀蛋白。色谱柱:Shimadzu,VP-ODS(5μm,150mm×4.6m);流动相:乙腈-水(35:65);流速1.0ml/min;柱温25℃;紫外检测波长303nm;大鼠静脉注射白藜芦醇20mg/kg后,于不同时间点取血测定血浆中药物浓度并估算其药物代谢动力学参数。结果白藜芦醇在0.023~23.600μg/ml浓度范围,其线性关系良好,r=0.9996,最低检测浓度约为0.007μg/ml。提取平均回收率为90.29%。大鼠静脉注射20mg/kg白藜芦醇后其消除半衰期为81.20min。结论HPLC灵敏度高,操作简便、准确,可用于大鼠血浆中白藜芦醇的测定及药物代谢动力学研究.Objective High performance liquid chromatography (HPLC) was established for detecting plasma concentrations of resveratrol and investigating its pharmacokinetics in rats with intravenous injection. Methods Plasma samples were treated with acetonitrile to deposit proteins. Shimadzu VP-ODS column (5 μm, 150 mm×4.6 mm) was used. The chromatographic conditions were as follows:The mobile phase was acetonitrile distilled water (35:65) ; the flow rate was 1.0 ml/min; 1he column temperature was at 25 ℃ ; and the ultraviolet detection wavelength was set at 303 nm. Rat blood samples were collected at different intervals after intravenous injection of a single dose (20 mg/kg) of resveratrol, and the concentration of resveratrol in rat plasma were determined by HPLC method for estimating pharmacokinetic parameters. Results There was a linear relationship of the concentration of resveratrol within the range of 0. 023 to 23. 600μg/ml (r=0. 999 6). The average recovery was 90.29%. The limit of detection of this method was 0. 007 μg/ml. The elimination half life was 81.20 min in rats with intravenous injection of 20 mg/kg resveratrol. Conclusion HPLC can be used for determination of resveratrol in rat plasma and pharmacokinetics study because of its high sensitivity, simplicity, accuracy and reliance.
