P16 P53在针刺乳腺增生模型大鼠乳腺组织的表达及意义

摘　　要：目的:通过观测乳腺增生大鼠及针刺治疗后乳腺组织中P16、P53蛋白表达的干预作用,探讨针刺对乳腺增生病的疗效与作用机理,为临床针刺治疗乳腺增生病,预防乳腺癌发生提供实验依据。方法:将48只健康雌性大白鼠随机分为4组,空白对照组、模型组、针刺治疗甲乙两组,每组12只。除空白对照组外,均采用贾氏造模方法建立乳腺增生模型。空白对照组、模型对照组的大鼠于第31天开始每日抓拿一次,不作任何治疗;针刺治疗的两组大鼠在第31天开始行穴位针刺治疗。治疗30天后处死大鼠,观测乳房形态大小,采集乳腺组织标本分别固定后,采用免疫组化法测定乳腺组织的P16、P53表达水平。结果:实验结果显示模型组造模成功,细胞结构中P16、P53表达明显;而针刺治疗组乳腺组织中P16、P53明显轻于模型组,两者比较有显著性差异。结论:针刺对乳腺增生大鼠乳腺组织P16、P53表达有干预作用,说明针刺对乳腺增生病有治疗作用,对乳腺癌发生有预防作用。Objective：By observing model rats with hyperplasia of mammary glands and the intervention effect expressed by P16 and P53 proteins in mammary tissue after acupuncture treatment,this paper discusses the curative effect and clinical effect of acupuncturing method on hyperplasia of mammary glands,thus in order to provide the experimental evidence for the treatment of hyperplasia of mammary glands clinically and the prevention of mammary cancer.Method：Divide 48 healthy female rats into four groups randomly and each group has 12 rats：blank control group,model group,acupuncturing treatment group A and B.All the rats should be built into models of hyperplasia of mammary glands by Jiashi method of model establishment except blank control group.From the 31st day,the rats of blank control group and model group will be caught one time each day without any treatment ;while the rats of acupuncturing treatment group A and B will be treated with acupuncturing.30 days later,observing the shape and size of died rats＇ breast and collecting the samples of mammary tissue,and then testing the expressing level of P16 and P53 in it by using immunohisochemistry.Result：Experiment result shows p16 and P53 are more obvious in cellular constructions of model group than p16 and P53 in cellular constructions of acupuncturing treatment group A and B.Conclusion：Acupuncturing treatment has intervention effect on the mammary tissue of the rats with hyperplasia of mammary glands.That is to say,acupuncturing treatment has a certain effect on hyperplasia of mammary glands and it also has a preventative effect on mammary cancer.
