摘　　要：目的观察肿瘤坏死因子α(TNF-α)在完全弗氏佐剂(CFA)致痛大鼠脊髓的表达。方法 SD大鼠56只,随机分为实验组(48只)和对照组(生理盐水组,8只)。实验组左足底皮下注射CFA。疼痛模型建立后1h、3h、1d、3d、7d、14d取大鼠胸、腰段脊髓,腰段与坐骨神经相连,用免疫组化和免疫荧光方法检测TNF-α表达。结果大鼠足底注射CFA后3h,同侧TNF-α表达较对侧和对照组增加(P<0.05);注射后3d,TNF-α表达最高;注射后14dTNF-α表达与对照组接近。TNF-α主要表达在神经元细胞。结论 TNF-α在CFA致痛大鼠脊髓后角的表达主要在炎性痛早期,并具有节段性,抑制TNF-α上调有可能成为临床治疗炎性痛早期的新方法。Objective To observe the expression of tumor necrosis factor（TNF-α） in the spinal cord following Complete Freund＇s adjuvant-induced inflammatory pain in rats. Methods Fifty-six SD rats were randomly divided into 2 groups of CFA（group CFA,48 rats） and normal saline（group C, 8 rats）. At 1 h,3, h, 1 d, 3 d, 7 d, 14 d after injecting CFA in left foot. Thoracic and lumbar segments of the spinal cord（the lumbar segments were contacted with the ischium nerve） were removed for detection of TNF-α with imrnunohistochemistry and immunofluorescence. Results The expression of TNF-α was higher in group CFA than that in the contralaterat group and group C on the 3rd hour after CFA injection（P〈0.05）, which reached peak level on the 3ra day and was comparable with that in group C on the 14th day. The expression of TNF-α was mostly seen in the neurons. Conclusion The expression of TNF-α increases mainly in the earlier stage after CFA induced-inflammatory pain with a characteristic of segments in rats. Clinically, suppression of the upregulation of TNF-α is possible in the future for improving inflammatory pain.
