健脾益气明目胶囊对MNU诱导的大鼠视网膜变性的保护作用

摘　　要：目的:观察健脾益气明目胶囊对N-甲基-N-亚硝脲(MNU)诱导的SD大鼠视网膜变性的保护作用。方法:出生后43d的雌性SD大鼠30只随机分为3组(n=10):正常对照组、模型对照组、药物组。药物组以健脾益气明目胶囊溶解后灌胃,正常组和模型组以等量纯化水灌胃,bid,连续7d。于生后50d,药物组和模型组大鼠腹腔注射MNU40mg.kg-1,正常组皮下注射等量生理盐水做对照。观察MNU处理后12,24,48,72h各组大鼠视网膜电图中a波及b波的变化。结果:正常对照组大鼠不同时间点视网膜电图(ERG)a波及b波的振幅差异无显著性,模型对照组在MNU处理后ERGa波及b波的振幅均呈持续性、进行性下降,健脾益气明目胶囊可明显抑制ERGa波及b波振幅的降低。结论:健脾益气明目胶囊对MNU诱导的大鼠视网膜变性有保护作用,是治疗视网膜变性性疾病的有效中药复方制剂。OBJECTIVE To observe the effects of Jianpiyiqimingmu capsule on N-methyl-N-nitrosourea （MNU） mdueed retinal degeneration in SD rats. METHODS 43-day female SD rats were randomly divided into： control group, model group, drug group. Jianpiyiqimingmu capsules were given i. g. to rats of drug treatment group for 7 days twice one day. Control group and model group were given with equal water. 50-day SD rats received a single intraperitoneal injection of 40 mg·kg^-1 MNU in model group and drug treatment group, while control group was given equal normal saline. Change of electroretinogram a wave and b wave were observed in 12, 24, 48, 72 hours after MNU injection. RESULTS The difference of the average amplitude of ERG a wave and b wave was not significant between different time in control group. The average amplitude of ERG a wave and b wave gradually decreased after MNU treatment in model group. Jianpiyiqimingmu capsule could evidently inhibit lose of a wave and b wave. CONCLUSION Jianpiyiqimingmu capsule has protective effects on MNU induced retinal degeneration in SD rats, and is an effective traditional Chinese compound preparation to treat the diseases of retinal degeneration.
