摘 要:采用PCR-RFLP(Polymerase chain reaction—restriction fragment length polymorphisma)分子标记技术,研究了292只滩羊KRT1.2(Keratin 1.2)基因的多态性及其与裘皮品质的关联性.检测出A和B两个等位基因,基因频率分别为85%和15%.产生AA,AB和BB 3种基因型,基因型频率分别为70.55%,28.42%和1.03%.最小二乘法分析表明,BB基因型所对应的毛股弯曲数最高,为6.33个,分别高于AA和AB基因型5.75个和5.67个.BB基因型所对应的毛纤维细度最高为37.52μm,分别高于AA和AB基因型37.04μm和36.36μm.但是,基因型AA,AB和BB之间均未达到显著差异(P>0.05).The polymorphism of KRT1.2 genes from 292 Tan sheep and their correlation with lamb fur qualities were analyzed with PCR-RFLP method.Two alleles A and B were detected with gene frequency of 85% and 15% respectively to generate three genotypes of AA,AB and BB with genotype frequencies of 70.55%,28.42% and 1.03% respectively.The results of least squares technique showed that the number of wool curvature corresponding to BB genotype was 6.33,which was significantly higher than that of 5.75 to AA genotype and 5.67 to AB genotype.The wool fineness corresponding to BB genotype was 37.52 μm,which was higher than that of 37.04 μm to AA genotype and 36.36 μm to AB genotype,but it was not significant between all genotypes(P0.05).
