摘 要:以骨形态发生蛋白受体IB(BMPR-IB)基因和促卵泡素β(FSHβ)亚基基因为小尾寒羊、陶滩寒二代、滩羊和无角陶赛特多胎性能候选基因,采用PCR-SSCP技术检测该基因在4个绵羊群体中的单核苷酸多态性,同时研究该基因对小尾寒羊高繁殖力的影响。结果表明:小尾寒羊及其杂种陶滩寒二代在BMPR-IB基因编码序列第746位碱基处发生了Q249R突变,而滩羊和无角陶赛特则没有发生这种突变;FSHβ基因CDS区外显子2在4个绵羊群体中都存在遗传多态性,CC基因型和DD基因型相比在外显子2第147处有1个T→C的单碱基突变,但未引起氨基酸改变。BMPR-IB的基因型和FSHβ的基因型分别在小尾寒羊及其杂种后代与滩羊和无角陶赛特之间分布差异极显著(P<0.01)或显著(P<0.05)。因此,BMPR-IB基因是影响小尾寒羊及其杂种陶滩寒二代高繁性能的一个主效基因,而FSHβ基因可能是控制小尾寒羊及其杂种陶滩寒二代高繁性能的一个主效基因或与之存在紧密连锁的一个遗传标记,均可用于对绵羊产羔数的辅助选择。Bone morphogenetic protein receptor IB(BMPR-IB) gene and follicle-stimulating hormoneβ-subunit gene (FSHβ) were selected as two fecundity candidate genes in Small-tailed Han sheep,its F_2 hybrids(Dorset×(Tan×Han), DTHF_2),Tan sheep and Poll Dorset sheep.The polymorphism of these two genes was studied by PCR-SSCP.The results indicated that the Q249R mutation of BMPR-IB gene as reported in Booroola Merino ewes existed in Small-tailed Han sheep and DTHF_2 sheep,whereas not present in Tan sheep and Poll Dorset sheep.Genetic polymorphisms of FSHβgene at exon2 in four sheep populations had been detected.There was one single nucleotide mutation(T^(147)→C^(147)) at exon 2 of FSHβgene in the CC genotype in comparison to the DD genotype,but this mutation did not cause amino acid change. The genotype distributions of BMPR-IB and FSHβwere significantly different(P0.01 or P 0.05) between high and low fecundity sheep populations.Therefore,these results demonstrated that BMPR-IB gene is a major gene that affects the prolificacy in Small-tailed Han and DTHF_2 sheep,and that the FSHβgene is either a major gene that influences the prolificacy in Small-tailed Han sheep and DTHF_2 sheep or a molecular marker in close linkage with such a gene,and could be used as two molecular genetic markers for improving litter size in sheep.
