摘  要:目的:运用比较蛋白质组学技术,探讨6周无负重游泳运动降低SHR大鼠血压血浆蛋白质表达的变化,以期进一步阐述运动治疗高血压病的机制。方法:以雄性4周龄SHR大鼠16只及WKY大鼠16只为研究对象,将其随机分为SHR运动组、SHR安静组、WKY安静组、WKY运动组。SHR运动组及WKY运动组进行1 h/天,5天/周,共6周的无负重游泳运动。使用CODATM2单通道无创血压测量仪检测老鼠血压,对处理好的血浆样品进行二维凝胶电泳和质谱分析。结果:(1)6周无负重游泳运动可以有效降低SHR大鼠的血压。(2)通过质谱分析和数据库检索,成功鉴定了6个差异蛋白:rCG45257、锚定蛋白重复域13家族成员D、甲状腺素转运蛋白A链、Ras相关域1家族同型A、胎球蛋白B前体和γ-肌动蛋白。Objective:Using the comparative proteomics method,the plasma protein expression was examined under the situation of blood pressure decreased by six-week load-free swimming training in spontaneously hypertensive rats(SHR)to further demonstrate the mechanism of hypertension improved by exercise.Methods:Sixteen male four-week old SHR and sixteen Wistar-Kyoto(WKY)rats were randomly divided into SHR exercise group,SHR rest group,WKY exercise group and WKY rest group.Rats in SHR exercise group and WKY exercise group exercised with a six-week load-free swimming protocol(1 hour per day,5 days per week).The blood pressure of rat was tested by CODATM2 single non-invasive blood pressure measurement appliance.The samples were used to do the two-dimensional gel electrophoresis and mass spectrometry.Results:The blood pressure was decreased significantly after six-week load-free swimming.The six proteins were identified by mass spectrometry and data base retrieval.They are rCG45257,similar to ankyrin repeat domain 13 family,member D,chain A,rat Transthyretin,Ras association domain family 1 isoform A,fetuin B precursor and γ-actin.
