STEAP4抗体干预对人前体脂肪细胞内活性氧、线粒体膜电位的影响

摘　　要：目的探讨STEAP4抗体干预对人前体脂肪细胞内活性氧(ROS)、线粒体膜电位的影响。方法应用锥虫蓝活性试验对细胞活性进行检测,并进一步确定STEAP4抗体原液按1:250的比例稀释。体外培养人前体脂肪细胞,应用质量浓度为0.5g.L-1的STEAP4抗体,按1:250的比例稀释后(即质量浓度为0.125g.L-1)干预人前体脂肪细胞。应用激光共聚焦显微镜拍照,定性、定量分析ROS、线粒体膜电位变化。结果 STEAP4抗体按1:250比例稀释作用于人前体脂肪细胞后,可显著降低细胞活性;人前体脂肪细胞在STEAP4抗体按1:250稀释浓度干预下,细胞内ROS水平显著增加、线粒体膜电位水平显著降低,差异均有统计学意义(Pa<0.05)。结论 STEAP4抗体按1:250比例稀释后,作用于人前体脂肪细胞后可显著提高ROS水平、降低其线粒体膜电位,推测STEAP4基因影响胰岛素敏感性的机制可能涉及线粒体功能变化。Objective To explore the effect of six-transmembrane epithelial antigen of prostate 4（STEAP4）antibody on the production of reactive oxygen species（ROS）and mitochondrial membrane potential in human pre-adipocytes.Methods Trypan blue test was used to determine the concentration of STEAP4 antibody.Human pre-adipocytes were cultured in vitro,1：250 concentration of the STEAP4 antibody was applied to interfer these cells.The STEAP4 antibody of 0.5 g·L-1 was used to be 1：250 concetration in order to interfer human pre-adipocytes.The ROS and the mitochondrial membrane potential changes were qualitatively and quantitatively analyzed by confocal laser microscopy.Results 1：250 STEAP4 antibodies（0.125 g·L-1）significantly reduced the activity of human pre-adipocytes.Intracellular ROS increased obviouslly,while mitochondrial membrane potential decreased significantly in the same condition（Pa0.05）.Conclusions Interfered by STEAP4 antibody,the intracellular ROS increased and the mitochondrial membrane potential reduced significantly.The affection of STEAP4 on insulin sensitivity might be related to the function of mitochondrial.
