摘  要:本文介绍纳金(nanogold,一种直径仅为1.4nm的超微金化合物)在包埋前单标和双标免疫电镜中的应用。电镜下观察到,以纳金标记的第7型代谢性谷氨酸受体(mGluR7)在大鼠脊髓后角浅层内定位于轴突终末,包括一些I-B4(一种从Grifonigsimplicifolig种子中得到的凝集素)标记的一级传入C纤维终末内的突触前膜上;在大鼠疑核内,μ阿片受体(MOR)主要存在于树突内的非突触部位。此外,我们还观察到,P物质(SP)与I-B4结合位点共存于一级传入C纤维中枢突终末内。实验证明,纳金标记的IgG或其Fab′片段是一种具有良好渗透性的免疫金探针,应用这种探针的包埋前免疫电镜术具有敏感性高、背景低、分辨率高等优点,尤其适于受体超微定位。e report an application of nanogold, an extremely small gold compound (1.4nm diameter), to singleor doublepreembedding immunoelectron microscopy. Under electron microscope, it was obesrved that, in the superficial laminae of the spinal dorsal horn of the rat, a metabotropic glutamate receptor, mGluR7, was localized in the presynaptic membranes of the axonal terminals some of which were the terminals of primary afferent C fibers labelled with isolectin IB4 from the seeds of Griffonia simplicifolia, and that opioid receptor (MOR) was mainly distributed in nonsynaptic sites of dendrites in rat ambiguous nucleus. It was also found that substance P(SP) was colocalized with IB4 binding afferent C fiber. From the practice it is demonstrated that IgG or its Fab fragment conjugated with nanogold is an immunogold probe which has the advantages of low background, high penetration, superior sensitivity and high resolutionan and sutis ultrastructural localization of receptors in particular when it is used in preembedding immunoelectron microscopy.
