摘  要:目的:探讨大承气颗粒对大鼠不完全性肠梗阻小肠上皮细胞的保护作用及机理。方法:采用贯穿缝扎末端回肠肠管的方法建立大鼠不完全性小肠梗阻模型,观察肠梗阻72h的小肠组织形态学、二胺氧化酶(DAO)活性、肠上皮细胞凋亡情况、小肠黏膜上皮细胞内Ca2+水平及血浆与小肠组织NO、MDA、SOD的变化。结果:模型组小肠黏膜损伤评分增加,血浆DAO活性明显升高,小肠组织DAO活性降低,肠组织细胞染色体DNA电泳可见典型的DNA梯状带生成,且小肠上皮细胞凋亡指数明显增加,肠上皮细胞内Ca2+浓度以及NO、MDA水平升高,SOD水平降低;大承气颗粒给药组不同程度地降低小肠黏膜损伤评分,降低血浆DAO及升高小肠组织DAO活性,降低小肠上皮细胞凋亡指数,且电泳呈现减弱的DNA梯状图谱、小肠黏膜上皮细胞内Ca2+浓度以及NO、MDA水平降低和SOD水平升高。结论:大承气颗粒能改善不完全性肠梗阻小肠组织形态学,阻遏肠黏膜上层绒毛中DAO的受损,抑制肠梗阻时的肠上皮细胞凋亡的过度增加,保护不完全性肠梗阻肠黏膜结构的完整性,具有肠屏障保护功能,上述作用机制可能与抗自由基及降低小肠黏膜上皮细胞内钙离子超载,从而减轻小肠黏膜上皮细胞损伤等有关。Objective:To study on the protective effects of Dachengqi granules against injury of IEC induced by incomplete intestinal obstruction in rats.Methods:Half of the terminal ileum of rat was ligated as incomplete intestinal obstruction model.Pathologic histology was observerd,apoptosis of enterocytes by agarose gel electrophoresis and apoptotic index by Hoechst staining were studied,levels of plasma and tissue DAO,NO,MDA,SOD,[Ca^2+]of IEC were detected.Results:Compared with the model group,grade of assessing histological mucosal damage decreased,plasma DAO decreased,while DAO in intestine increased;apoptotic index decreased and it was also confirmed by DNA ladder formation;[Ca^2+]of IEC decreased;NO and MDA of plasma and intestine both decreased,but SOD of those increased.Conclusion:Dachengqi granules can improve mucosal morphology,inhibit the release of DAO and excess enterocytes apoptosis,lessen IEC injury and protect IEC to enhance the intestinal barrier function.The mechanisms above may be concerned with anti-free radicals,decreasing [Ca^2+]of IEC to lessen injury to IEC.
