摘  要:目的:观察清风胶囊对吗啡依赖小鼠反复染毒后自主活动、耐力及血中c-fos基因表达的影响。方法:按剂量递增法建立吗啡依赖小鼠模型,经纳洛酮催促戒断后,停药7天,再按上述方法重复给药及戒断两次。模型建立后各组小鼠灌胃给予相应药物治疗5天。采用多功能小鼠自主活动记录仪观察小鼠的自主活动次数,采用小鼠游泳试验观察小鼠的游泳时间,采用免疫组化法检测小鼠血中c-fos基因表达。结果:与空白对照组比较,吗啡模型组小鼠自主活动次数明显减少,游泳时间显著缩短,血中c-fos阳性细胞数明显增多。给予清风胶囊治疗后,小鼠自主活动次数明显增多,游泳时间显著延长,血中c-fos阳性细胞数趋于正常,其中清风胶囊高、中剂量组的作用显著。结论:清风胶囊能提高反复成瘾戒断小鼠的活力和耐力,调节染毒小鼠血中c-fos基因的表达。Objective:To investigate the effects of Qingfeng Capsules on locomotor activity,swimming time and c-fos gene expression of blood in the morphine-dependent mice,and to discuss the TCM mechanism of Qingfeng Capsules treating protracted withdrawal syndrome.Methods:Morphine-dependent mouse model was established by a gradually increasing doses and repeated the processes of morphine addiction-withdrawal two times.The mice were treated with Qingfeng Capsules for 5 days,locomotor activity and swimming time of mice were evaluated by a multi-functional determinator for little animals and a mouse swimming test.The c-fos gene expression was detected with the immunohistochemical method.Results:In morphine-dependent model group,frequency of locomotor activity in mice was markedly decreased,swimming time of mice was significantly shortened and the c-fos positive cell counts in mouse blood was obviously increased.In three doses of Qingfeng Capsule groups,after the mice were treated with Qingfeng Capsules,frequency of locomotor activity and swimming time of mice were significantly increased and the c-fos positive cell counts was obviously reduced,in which the effects of high-dose and mid-dose of Qingfeng Capsules were more marked.Conclusion:Qingfeng Capsules could enhance locomotor activity and swimming time and regulated the c-fos expression in mice those were exposed to morphine repeatedly.
