摘 要:目的研究怡肾汤对肾间质纤维化大鼠TGF-β1和ColⅢ表达的影响,探讨其抗肾间质纤维化的作用机制。方法 32只雄性SD大鼠随机分为正常对照组、模型组、怡肾汤组和依那普利组,每组8只。适应性饲养1周后,除正常对照组外其余均采用腺嘌呤灌胃法建立大鼠肾间质纤维化动物模型。第1、2、3周末采血分离血清测定肌酐、尿素氮含量。第21天处死大鼠,分离肾脏,应用HE及Masson染色,观察肾脏组织病理学改变,免疫组化方法检测肾组织转化生长因子-β1(TGF-β1)和Ⅲ型胶原(ColⅢ)的表达,结果应用图像分析系统进行半定量分析。结果模型组、怡肾汤组及依那普利组与正常组比较均呈现程度不同的病理损害。免疫组化半定量分析显示怡肾汤组肾间质TGF-β1、ColⅢ的表达量均少于模型组(P<0.05)。此外,怡肾汤还能降低血BUN、Scr水平。结论怡肾汤可能通过下调TGF-β1、ColⅢ水平,减少ECM沉积,从而达到防止肾间质纤维化的作用。Objective To investigate the possible preventive mechanism of "Yishen Decoction" in the relief of renal interstitial fibrosis in terms of expressions of TGF-β1 and ColⅢ in rats.Methods Thirty-two male SD rats were randomly divided into 4 groups: normal group,model group,Yishen Decoction group and Enalapril group.The models of renal interstitial fibrosis were induced by intragastric administration of adenine.At the end of 1st,2nd and 3rd weeks,the serum Scr and BUN levels were checked;then the rats were sacrificed and the renal tissues were taken for pathological examination by HE staining and Masson staining;immunohistochemistry was used to measure the expressions of TGF-β1 and collagen Ⅲ(ColⅢ) in nephridial tissue.Results In the model group,Yishen Decoction group and Enalapril group there were kidney lesions in different degrees as compared with the normal group.The expressions of TGF-β1 and collagen Ⅲ(ColⅢ) in Yishen Decoction group was lower than those in the model group(P〈0.05);and Yishen Decoction could significantly decrease the serum levels of BUN and Scr.Conclusion "Yishen Decoction" can relieve renal interstitial fibrosis by decreasing TGF-β1 and ColⅢ levels and reducing ECM deposit.
