区带毛细管电泳法同时测定连翘不同部位中5个木脂素类成分的含量

摘　　要：目的:建立同时测定连翘中连翘苷、连翘脂素、(+)-表松脂素-4′-O-葡萄糖苷、(+)-松脂素-β-D-葡萄糖苷和(+)-表松脂素5个木脂素类成分的区带毛细管电泳法,并测定连翘不同部位中5个成分的含量。方法:石英毛细管柱(75μm×60cm,有效长度30cm)作为分离通道,以50mmol·L-1硼砂(用0.1mol·L-1氢氧化钠溶液调pH至9.90)为电泳缓冲液,工作电压15kV,温度20℃,检测波长214nm。结果:5个成分在12.5min内实现良好分离,连翘苷、连翘脂素、(+)-表松脂素-4′-O-葡萄糖苷、(+)-松脂素-β-D-葡萄糖苷和(+)-表松脂素浓度分别在3.6～145μg·mL-1、0.5～20μg·mL-1、0.68～27.2μg·mL-1、6.8～272μg·mL-1和0.6～24.2μg·mL-1范围内呈良好的线性关系(r≥0.9985);平均回收率(n=5)分别为98.50%,101.2%,100.4%,97.36%,102.0%。结论:该方法简便、快速、准确,为科学评价与有效控制连翘药材质量提供新方法。连翘叶和花中连翘苷和(+)-松脂素-β-D-葡萄糖苷的含量均高于果实。Objective：To develop a capillary zone electrophoresis（CZE） method for the determination of phillyrin,phillygenin,（＋）-epipinoresinol-4＇-O-glucoside,（＋）-pinoresinol-β-D-glucoside and （＋）-epipinoresinol in different parts of Forsythia suspensa. Methods：Capillary electrophoresis was performed using a fused silica capillary column（75 μm×60 cm,an effective length of 30 cm）. The running buffer was composed of 50 mmol·L-1 borax （adjust to pH 9.90 with 0.1 mol·L-1 sodium hydroxide solution）. The applied voltage was 15 kV and the temperature was 20 ℃.The detection wavelength was 214 nm.Results：The five components were separated successfully within 12.5 min;A good linearity （r≥0.9985） was found in the ranges of 3.6-145,0.5-20,0.68-27.2,6.8-272 and 0.6-24.2 μg·mL-1 for the five components,respectively;The mean recoveries（n=5） were 98.50%,101.2%,100.4%,97.36% and 102.0%.Conclusion：The operation of the method is simple,quick,accurate and can be used as a new method for the quality control of Forsythia suspensa.The contents of phillyrin and （＋）-pinoresinol-β-D-glucoside in leaf and flower are much higher than those in fruit.
