摘  要:目的:建立天麻中对羟基苯甲酸和4,4′-二羟基二苄砜的含量测定方法。方法:采用HPLC法,使用KromasilC18(250mm×4.6mm,5μm)色谱柱,以甲醇-0.1%醋酸水溶液为流动相,梯度洗脱,流速1.0mL.min-1,检测波长240nm,柱温35℃。结果:对羟基苯甲酸和4,4′-二羟基二苄砜进样浓度分别在0.421~8.41μg·mL-1(r=0.9992)和0.650~13.0μg.mL-1(r=0.9999)范围内与峰面积线性关系良好;平均加样回收率(n=9)分别为98.2%和98.7%。结论:采用HPLC法测定天麻中对羟基苯甲酸和4,4′-二羟基二苄砜的含量方法准确可靠,可用于天麻的质量控制。Objective:To establish a reversed-phase HPLC method for simultaneous determination of p-hydroxybenzoic acid and 4,4'-dihydroxybenzyl sulfone in the rhizome of Gastrodia elata Bl..Methods:A Kromasil C18(250 mm×4.6 mm,5 μm) column was adopted with a methanol-water-0.1% acetic acid gradient elution system.The elution were detected at 240 nm,the flow rate was 1.0 mL·min-1 and the column temperature was 35 ℃.Results:The linear range of p-hydroxybenzoic acid and 4,4'-dihydroxybenzyl sulfone were 0.421-8.41 μg·mL-1(r=0.9992),0.650-13.0 μg·mL-1(r=0.9999),respectively.The average recoveries(n=9) of the two components were 98.2%,98.7%,respectively.Conclusion:The method is accurate,sensitive and reliable for determination of p-hydroxybenzoic acid and 4,4'-dihydroxybenzyl sulfone in the rhizome of G. elata. And this method can be used for the quality control research of Gastrodia elata Bl..
