摘  要:目的:建立高效液相色谱法同时测定布渣叶中牡荆苷和异鼠李素-3-O-β-D-芸香糖苷含量。方法:采用KromasilC18色谱柱(250mm×4.6mm,5μm),流动相为甲醇-0.1%磷酸水溶液,二元梯度洗脱,流速1.0mL.min-1,检测波长276nm,柱温35℃。结果:牡荆苷和异鼠李素-3-O-β-D-芸香糖苷的线性范围分别为0.0412~0.412μg(r=0.9992)和0.0424~0.424μg(r=0.9991);平均回收率(n=9)分别为96.2%和96.0%。结论:该方法简便、准确,可用于布渣叶中牡荆苷和异鼠李素-3-O-β-D-芸香糖苷含量的同时测定。Objective:To develop an RP-HPLC method for the simultaneous analysis of vitexin and isorhamnetin-3-O-β-D-rutinoside in Microcos paniculata L.. Methods:HPLC determination was performed on a Kromasil C18 column(250 mm×4.6 mm,5 μm)with a binary elution of methanol-0.1 % phosphoric acid aqueous solution as mobile phase and detected at 276 nm.The flow rate was 1.0 mL·min-1 and the column temperature was 35 ℃. Results:The calibration curves were linear in the ranges of 0.0412-0.412 μg(r=0.9992)and 0.0424-0.424 μg(r=0.9991)for vitexin and isorhamnetin-3-O-β-D-rutinoside,and the average recoveries(n=9)of vitexin and isorhamnetin-3-O-β-D-rutinoside were 96.2% and 96.0%,respectively.Conclusion:This method is simple and accurate,which can be used for the simultaneous determination of vitexin and isorhamnetin-3-O-β-D-rutinoside in Microcos paniculata.
