摘　　要：的探讨骨调素（ｏｓｔｅｏｐｏｎｔｉｎ，ＯＰＮ）在系统性红斑狼疮（ＳＬＥ）患者中的表达及临床意义。方法用流式细胞仪和Ｎｏｒｔｈｅｒｎｂｌｏｔ杂交方法分别检测ＳＬＥ患者外周血单个核细胞（ＰＢＭＣ）ＯＰＮ蛋白和ｍＲＮＡ的表达。结果活动期和稳定期ＳＬＥ患者ＰＢＭＣＯＰＮ蛋白和ｍＲＮＡ的表达均高于正常人（Ｐ＜００１），ＳＬＥ患者ＯＰＮ蛋白主要在淋巴细胞表达，而活动期患者ＰＢＭＣＯＰＮ蛋白及ｍＲＮＡ的表达高于稳定期患者（Ｐ＜００５）。结论ＳＬＥ患者ＰＢＭＣＯＰＮ表达增高，提示ＯＰＮ在人类ＳＬＥ的发病机制中有重要作用，ＯＰＮ可能作为ＳＬＥ活动性指标之一。Objective To investigate osteopontin(OPN)expression in SLE patients and its clinical significance. Methods The OPN protein and mRNA expression in peripheral blood mononuclear cells (PBMC) from 80 patients and 30 healthy persons was detected by cytometry and Northern blot analysis method. Results The OPN protein and mRNA expression of PBMC in SLE patients were significantly higher than in controls (P<001).OPN protein expressin of lymphocyte was significantly higher than that of monocyte in SLE patients.There were significant differences in PBMC protein and mRNA expression between active group and inactive group (P<005). Conclusion OPN abnormal expression in SLE patients suggests that OPN may play an important role in the pathogenesis of SLE.OPN may be considered as a marker for estimating activity of SLE.
