不同浓度的华蟾素对卵巢癌3AO细胞MMP-2和TIMP-2表达的影响及可行性研究

摘　　要：目的探讨华蟾素对卵巢3AO细胞基质金属蛋白酶(MMP-2)和组织抑制因子(TIMP-2)表达的影响及其临床意义。方法体外培养卵巢癌3AO细胞,不同浓度的华蟾素干预后,用SABC免疫组化法检测MMP-2和TIMP-2表达。结果卵巢癌3AO细胞可表达MMP-2和TIMP;0.25mg/ml华蟾素对卵巢3AO细胞MMP-2和TIMP-2的表达无显著影响(P>0.05);2.5mg/ml华蟾素干预后,卵巢癌3AO细胞MMP-2的表达显著下降,TIMP-2的表达显著升高(P<0.05);25mg/ml与2.5mg/ml华蟾素对卵巢癌3AO细胞MMP-2和TIMP-2表达影响无统计学差异(P>0.05)。结论 2.5mg/ml华蟾素可显著降调卵巢癌3AO细胞MMP-2的表达,升调TIMP-2的表达,两者的失衡可抑制卵巢癌3AO细胞的侵袭性生长。Objective To investigate the effect and clinical significance of cinobufacine on the expression of MMP-2 and TIMP-2 in the ovarian carcinoma 3AO ceils cultured in vitro. Methods Ovarian carcinoma 3AO ceils were cultured in vitro; after the action of cinobufacine with different concentration, the expression of MMP-2 and TIMP-2 was measured by SABC immunohistometry technigue. Restflts The olarian carcinoma 3AO cells caltured in vitro could express MMP-2 and TIMP-2; 0.25mg/ml cimobufacine didn＇t affect the expression of MMP-2 and TIMP-2 in the avarian carcimoma 3AO cells （P〉0.05）; after the action of 2.5mg/ml cimobufacine, the expression of MMP-2 was down regulated and the TIMP-2 was unregulated significantly （P〉0.05）; the action of 25mg/ml cinobufacine on the MMP-2 and TIMP-2 was similar to that of 0.25mg/ml cinobufacine （P〉0.05）. Coclusion 2.5mg/ml cimobufacine can down regulate the expression of MMP-2 and up regulate the expression of TIMP-2. The imbalance of MMP-2 and TIMP-2 may inhibit the invasive grouth of ovarian carcinoma 3AO cells.
