摘  要:目的探讨分泌的卷曲相关蛋白2(SFRP2)在耳垂瘢痕疙瘩发生发展过程中的表达及作用,为瘢痕疙瘩的基因治疗提供新的线索。方法利用原位杂交和WesternBlot检测伤后3、6、12、24个月耳垂瘢痕疙瘩组织边缘区和中央区SFRP2的表达变化。结果在耳垂瘢痕疙瘩组织中,边缘区伤后12个月组SFRP2的表达显著高于3、6个月组(P〈0.01),而24与12个月组比较,差异无统计学意义。中央区伤后12个月SFRP2的表达出现下降(P〈0.01);边缘区SFRP2的表达均高于同一时期中央区的表达(P〈0.05,P〈0.01)。瘢痕疙瘩表皮和血管内皮及周围组织SFRP2的表达也增强。SFRP2mRNA和蛋白质的变化相似,只是中央区SFRP2蛋白质的衰减较SFRP2mRNA延后。结论SFRP2的高表达可能参予了瘢痕疙瘩的生长发展,而且瘢痕疙瘩表皮及边缘区SFRP2的高表达对其浸润性、持续性生长作用强于中央区。血管内皮细胞及周围组织SFRP2的高表达也发挥了作用。降低SFRP2的表达可能是基因治疗瘢痕疙瘩的新途径。Objective To investigate the expression and the role of secreted frizzled-related protein 2 (SFRP2) in the earlobe keloid and find a valid way to treat the keloid with gene therapy. Methods The expression of SFRP2 mRNA and protein was tested with in situ hybridization and Western Blot Analysis method in the different period of earlobe keloid. Results The SFRP2 mRNA and protein expression at the keloid edge was significantly high in 12 month group than in 3 or 6 month groups(P 〈 0.01 ) , but not than in 24 month group. The SFRP2 expression started to decrease in the keloid center 12 month later ( P 〈 0. O1 ). The SFRP2 expression was always higher in edge than in center during all the period( P 〈 O. 05, P 〈 O. 01 ). Conclusions The results suggest that SFRP2 may play an important role in the development of keloid, especially at the keloid edge. The high SFRP2 expression in endothlial cells and surrounding tissue is also important. It may be a new way for gene therapy of keloid by decreasing the SFRP2 expression.
