摘  要:目的 探讨紫杉醇对血管紧张素Ⅱ(Ang Ⅱ)诱导的大鼠降主动脉外膜成纤维细胞(AFs)迁移能力的影响.方法 采用组织帖块法原代培养AFs并传代,取传至4~6代的AFs用于实验.分别以0、10、30、90nmol/L的紫杉醇干预12h,确定对AFs生存活力无影响的紫杉醇浓度.取AFs分为Ang Ⅱ组及Ang Ⅱ+9nmol/L紫杉醇组,采用Transwell小室测试细胞迁移能力,并采用Western bloting检测两组MMP-2的表达量.作比较分析.结果 10nmol/L以内浓度的紫杉醇均不会影响成纤维细胞的生存活力.0、9nmol/L紫杉醇组成纤维细胞迁移能力分别为0.56±0.079、0.44±0.047,9nmol/L紫杉醇组较0nmol/L紫杉醇组的侵袭能力明显下降(P〈0.01 o 9nmol/L紫杉醇组MMP-2的表达量明显低于0nmol/L紫杉醇组(P〈0.05),其降幅可达到24.0%.结论 紫杉醇可抑制Ang Ⅱ诱导的大鼠AFs的迁移,可能与抑制MMP-2的表达有关.Objective To investigate the effect of taxol on the migration of adventitial fibroblast cells (AFs) induced by angiotensin Ⅱ(Ang Ⅱ). Methods AFs were prepared from rat aortas and the cells of 4th - 6th passage were used in the study. AFs were treated with Ang Ⅱ or AngⅡ +9nmol/l taxol. Cell migration was determined by Transwell Chamber. The expression of MMP-2 was determined by Western blotting. Results AFs migrations in Ang Ⅱ group and Ang Ⅱ +9nmol/I taxol were 0.56+0.079 and 0.44+0.047 respectively (P〈0.01). Western blot indicated that compared with Ang Ⅱ group, the expression of MMP-2 in Angll + 9 nmol/l taxol group decreased by 24% (P〈0.05). Conclusion Taxol can effectively inhibit the migration of AFs,which may be associated with down-regulation of MMP-2.
