摘　　要：目的 探讨系统性红斑狼疮（SLE）患者外周血单个核细胞中Brn-3a mRNA 表达及其与疾病活动性的关系.方法 以β-actin为内参照,建立逆转录实时荧光定量聚合酶链反应（FQ-RT-PCR）方法,在Light Cycler Real time PCR检测仪上检测92例SLE患者（活动期46例,缓解期46例）、其他结缔组织病（CTD）患者30例（疾病对照组）、健康体检者30例（正常对照组）外周血单个核细胞（PBMCs）中Brn-3a mRNA 的表达水平和含量.以ΔCt=Ct（待测基因）-Ct（内参基因）来比较基因表达水平的高低.结果 活动期SLE患者Brn-3a mRNA表达水平明显高于缓解期SLE患者（P〈0.01）;活动期SLE患者Brn-3a mRNA表达水平明显高于正常对照组与疾病对照组（P〈0.01）;缓解期SLE患者Brn-3a mRNA表达水平与正常人及疾病对照组差异无统计学显著性意义（P〉0.05）.结论 活动期SLE患者Brn-3a mRNA表达水平显著高于正常人、疾病对照组和疾病缓解期,提示Brn-3a的表达与SLE的发生发展存在一定关联性,可能参与了SLE的发病过程并与疾病的活动性有关.Objective To evaluate the expression of Brn-3a mRNA on peripheral blood mononuclear cells （PBMCs） in patients with systemic lupus erythematosus（CSLE）. To find out the relationship between Brn-3a and disease activity. Methods The expression of Brn-3a mRNA were detected by reverse transcriptlon-reat time quantitative polymerase chain reac- tion （FQ-RT-PCR） on PBMCs in 92 patients with SLE（46 active patients,46 remittent patients）,30 patients with other rheumatic diseases as diseases controls,and 30 health controls. β-aclin were used as endogenous control. The results were calculated by the formula △Ct（objective gene）-Cl（endogenous reference gene）. Results The values of Brn-3a mRNA expression in active patients were higher than that in remittent patients（P〈0.01）,patients with other rheumatic discases （P 〈 0. 01） and healthy controls （P〈0. 01）. There were no significant difference among remittent patlents,other rheumatic diseases patients and healthy controls（P〈0.05）. Conclusion The study indicates that the levels of Brn-3a mRNA expression on PBMCs is significantly elevated in patients with active SLE,and it may be involved in pathogenesis of disease.
