慢性乙肝患者血清趋化因子MCP-1浓度与病毒载量关系的研究

摘　　要：目的 研究慢性乙型肝炎（CHB）患者外周血清中单核细胞趋化蛋白-1（MCP-1）的表达水平与HBV DNA载量之间的相关性.方法 用ELISA检测CHB患者和健康对照者血清中的MCP-1 蛋白浓度,以实时荧光定量PCR检测CHB患者血清中HBV DNA拷贝数.结果 CHB组血清中MCP-1浓度是157±80 ng/L,健康对照组MCP-1浓度是265±44 ng/L,CHB组血清中MCP-1浓度显著低于健康对照组,t=5.355,P＜0.001,差异有统计学显著性意义;CHB患者血清中MCP-1浓度与血清中HBV DNA拷贝数呈负相关（r=-0.602,P＜0.001）.CHB患者血清中MCP-1的浓度是降低的,MCP-1浓度与血清中HBV DNA 载量之间呈负相关（r=-0.602,P＜0.001）.结论 乙肝病毒与宿主细胞相互作用下调机体MCP-1表达,可能对HBV致病机理的研究提供一个新的视点.Objective To investigate expression level of monocyte chemoattractant pretein -1 （MCP-1） and c,Srrelation between the concentration of MCP-1 with HBV DNA load in serum of chronic hepatitis B（CHB） patients. Methods The ELISA was applied for detecting the concentration of MCP-1 in serum of CHB patients and control groups. Real-time fluorescence quantitative polymerase chain reaction assay was used to analyse HBV DNA copies in the serum of CHB patients. Results The concentration of MCP-1 in CHB patients＇ serum was 157±80 ng/L,and that of in the control group was 265 ± 44 ng/L. There were significantly low in CHB patients （t= 5. 355 ,P〈0. 001）. Significantly negative correlation lied between the concentration of MCP-1 and DNA copies in the CHB patients＇serum （r=-0. 602,P〈0.01）, The expression of MCP-1 was down-regulated in serum of CHB patients,negative correlation lies between the concentration of MCP-1 and the HBV DNA load in CHB patients＇ serum. Conclusion These suggest that there may be interaction between viral proteins with host cells which affect the expression of MCP-1.
