摘  要:目的观察内皮素-1(endothelin-1,ET-1)在实验性视网膜脱离兔中的表达。方法选取18只(36眼)中国大白兔,对照组2只(4眼);实验组16只(32眼)。实验组大白兔在间接检眼镜直视下,用50nm玻璃微管在兔眼下方600钟位赤道部视网膜造孔并注射0.5mL生理盐水于视网膜下间隙。实验组各取2只4眼兔于术后0.5h、1h、3h、6h、24h和3d、7d、14d摘除眼球作组织学观察并以免疫组织化学和原位杂交的方法检测ET-1在视网膜的表达,并于抽取玻璃体降眼压以及取眼球时分别抽出玻璃体用放射免疫测定法检测ET-1浓度。结果实验组兔眼术后均发生视网膜脱离,视网膜脱离范围300-900钟位。从模型建立后3h,经放射免疫测定开始可检测出玻璃体中存在ET-1,浓度逐渐上升,并随后下降,而免疫组织化学的结果提示,ET-1在模型建立后24h开始表达。原位杂交检测结果也证实,建模后玻璃体液中检测出ET-1,ET-1的mRNA主要表达在视网膜神经层各层细胞和RPE的细胞核,胞浆只有较弱的着色,造模24h后,胞浆的着色才明显加深,并且在建模后3d时着色明显,7d、14d减弱。结论 ET-1在视网膜脱离中是一种早期表达的细胞因子,有峰值,也有低谷,除与视网膜脱离的损伤有关外,其在视网膜下液含量的波动与外屏障的破坏和细胞因子间的相互作用有关。Objective To investigate the expression of endothelin-1(ET-1)in rabbits with experimental retinal detachment.Methods Eighteen Chinese albino rabbits(36 eyes)were divided into control group(2 rabbits,4 eyes)and experimental group(16 rabbits,32 eyes).Under the indirect ophthalmoscope,rabbits in experimental group were made a hole of retina at 6 o'clock under ambitus,and injected with 0.5 mL nature saline into the subretinal space by a glass-made pipette with a flat tip of 50 nm diameter.The eyeballs of 4 eyes(2 rabbits)in experimental group were taken out to make histological observation at postoperative half an hour,1 hour,3 hours,6 hours,24 hours,3 days,7 days and 14 days,respectively.The expression of ET-1 in retinas was measured by immunohistochemistry and situ hybridization.After drawing out vitreous body,lowering intraocular pressure and taking out eye ball,concentration of ET-1 was detected by radioimmunoassay.Results The retinal detachment showed in all 32 eyes of rabbits in experimental group,the detachment scope was from 3 to 9 o'clock.The concentration of ET-1 in vitreous body was detected at 3 hours after modeling,after gradually increase,there was a decrease.Immunohistochemical results showed that the concentration of ET-1 was found at 24 hours after modeling,while 3 hours after modeling by situ hybridization.Expression of ET-1 mRNA was found in each nervous layer of retina and RPE nucleus,and there was a light coloring in endochylema.Coloring of endochylema was gradually deepened at 24 hours after modeling,more obvious at 3 days after modeling and decreased at 7 days and 14 days after modeling.Conclusions ET-1 is a kind of earlier expressed cytokine in retinal detachment.Its expression has peak and low.Besides of lesion by retinal detachment,its fluctuation in subretinal fluid has relation with the damage of external shield and correlation among cytokines.
