摘 要:目的研究早熟凝集染色体(PCC)技术在核事故中快速估算受照剂量的方法,以备突发事故应急之用。方法改进常规的PCC方法;以改进后的方法研究苦杏仁苷和细胞松驰素B(Cyt-B)在60Coγ射线大剂量照射下对促进人血淋巴细胞的增殖作用;从中选择最优化的实验方法,并用此方法建立剂量-效应曲线和验证。结果①PCC方法的改进:以花萼海绵体诱癌素A(CA)35 nmol/L、秋水仙素30 nmol/L培养50 h后收获的细胞形态最好,可用于分析的细胞比例最高;②受照0~20.0 Gy后,以苦杏仁苷为150 mg/L和Cyt-B为0.2 mg/L共同培养收获的细胞增殖率最高;③受照10.0、15.0、20.0 Gy后以最佳增殖浓度培养44 h的细胞增殖率相当常规法56 h培养的结果;④以最佳增殖浓度、52h培养建立0~20.0 Gy的剂量-效应曲线,受照剂量与PCC环的相关系数为0.997 7(P<0.01),直线回归方程为y^=0.070 9x-0.040 7,相关指数为0.995 5;⑤3个验证样本(5.0、10.0、20.0 Gy)读片结果用本研究的标准曲线推算出的剂量相当原设定剂量的96.0%~105.9%。结论本研究选用的2种细胞增殖促进剂在60Coγ射线0~20.0 Gy的剂量受照下具有促进细胞快速增殖的作用,建立的剂量-效应曲线相关指数高,适用于核和电离辐射事故时快速估算受照生物剂量。Objective To study the technology of premature chromosome condensation(PCC) for quickly estimating radiation dose in nuclear accident emergency.Methods The improved conventional PCC method was used to study the proliferation effect of 2 kinds of cell proliferation promoting agents,amygdalin and Cyt-B,on human peripheral blood lymphocytes after high dose ionizing radiation with 60Co γ-ray.The optimum proliferation concentration and optimum cultivation time was chosen to establish the 0-20.0 Gy dose curve of 60Co γ-ray.Results ①Based on the improved PCC technology,calyculin A(CA,35 nmol/L) and colchicines(30 nmol/L) were added and cultivated for 50 hours.The cells with best morphology and structure could be harvested for further observation.The proportion of analyzable G2+M period cells was the highest(70.3%).②After being irradiated by 0-20.0 Gy 60Co γ-ray,and the cells were cultivated for 50 hours with amygdalin(150 mg/L) and Cyt-B(0.2 mg/L),the cell proliferation rate was the highest,and the chromosome rings which were the analysis index of aberration were clear and recognizable.③With the optimum proliferation condition,the cell proliferation rate which irradiated at 10.0,15.0 and 20.0 Gy for 44 hours were the same as the conventional 56-hour cultivation.With 48 to 52 hours cultivation,it could harvest enough analyzable cells which irradiated at 20.0 Gy.④The dose curve of irradiating at 0-20.0 Gy 60Co γ-ray was established by the optimum proliferation condition of 52-hour cultivation.The correlation coefficient of radiation dose and chromosome ring was 0.997 7(P0.01).⑤With our optimum method,the samples irradiated by 60Co γ-ray from another laboratory were cultivated.Based on 3 irradiated spots(5.0,10.0 and 20.0 Gy),the doses calculated by our proposed standard curve were close to the setting dose(96.0%-105.9%).Conclusion Two of the cell proliferation promoting agents play a role in the promotion of rapid cell proliferation.The dose-effect curve has a high corr
