泽泻汤对代谢综合征大鼠胰岛NPY及其Y1R影响的研究

摘　　要：目的:观察泽泻汤对代谢综合征(MS)大鼠血浆NPY,胰岛NPY及其Y1R表达的影响,探讨其可能的治疗机制。方法:用高糖高脂饲料喂饲建立MS大鼠,将23只MS大鼠分为三组,分别给盐水、泽泻汤、西布曲明。4周后测体重、血糖、TG,用放免法检测血清胰岛素、血浆NPY;免疫组化法检测胰岛NPY及其Y1R表达,用im age-pro p lus 6.0进行平均光密度分析。结果:与盐水组相比,泽泻汤组大鼠体重、血糖、TG、血清胰岛素、血浆NPY水平、胰岛NPY及其Y1R表达的平均光密度均显著降低(P<0.05)。结论:泽泻汤对MS大鼠有良好的治疗作用,其作用机制可能与抑制胰岛NPY及其Y1R表达有关。Objectives：To observe the effect of Zexie Decoction on plasma NPY and pancreatic islets NPY Y1 receptor subtype（NPYY1R,Y1R）in metabolic syndrome rats and the possible mechanism.Methods：Developping models of Metabolic Syndrome rats by feeding high sugar fat diet.Twenty-three MS rats were randomly divided into saltwater group,TCM（Zexie Decoction）group and Western Medicine（Sibutramin）group.Body weight,GLU,TG,Serum insulin and Plasma NPY of the rats were measured after four weeks.Meanwhile,NPY Y1R in pancreatic islets were tested by immunohistochemistry.The density mean was analyzed by image-pro plus 6.0.Results：Zexie Decoction could inhibit the growth of body weight and reduce the levels of GLU,TG,serum insulin and plasma NPY（P〈0.05）.NPY Y1R in pancreatic islets were significantly lower in TCM group than those in the saltwater group（P〈0.05）.Conclusions：There is obvious therapeutic effect of Zexie Decoction on metabolic syndrome rats,and the mechanism is possibly which.related to its inhibition of the synthesis of NPY and the expression of Y1R in pancreatic islets.
