半定量RT-PCR法检测VCP mRNA与PCNA mRNA在胃癌中的表达

摘　　要：目的通过研究癌旁正常胃组织、淋巴结转移癌组织及胃癌组织中VCP mRNA的表达及其与增殖细胞核抗原(PCNA)mRNA表达的相关性,探讨VCP mRNA在胃癌发生发展过程中的可能作用,为胃癌的诊断及预后判断提供分子生物学指标。方法运用半定量RT-PCR技术检测42例胃癌患者胃癌组织、淋巴结转移癌组织、相应癌旁正常胃组织中VCP mRNA、PCNA mRNA的表达水平。结果 (1)VCP mRNA、PCNA mRNA在胃癌组织、淋巴结转移癌组织较相应癌旁正常组织表达显著升高(P<0.01),两者在胃癌组织中的表达丰度与肿瘤的分化、浸润深度、淋巴结转移个数及病理TNM分期有相关性(P<0.05);(2)VCP mRNA、PCNA mRNA在胃癌组织中表达存在正相关关系(r=0.631,P<0.01)。结论胃癌组织中VCP mRNA高表达可能是上调PCNA mRNA表达的一个因素,对于肿瘤的发生发展具有重要影响,联合检测两分子指标的表达对预测胃癌患者预后?指导治疗有一定临床辅助意义。ObjectiveTo study expressions of valosin containing protein（VCP） mRNA of gastric cancer,lymphatic invasive cancer and corresponding adjacent normal gastric tissue and its correlation with proliferating cell nuclear antigen（PCNA） mRNA expression of gastric cancer,and investigate the possible roles of VCP mRNA in the gastric carcinogenesis and progression,so as to provide the new molecular biological basis on diagnosis and prognosis assessment of gastric cancer.Methods Expressions of VCP mRNA and PCNA mRNA were dectected by semi-quantitative reverse transcriptase polymerise chain reaction（RT-PCT） in 42 cases of gastric cancer,lymphatic invasive cancer and corresponding adjacent normal gastric tissue.Results（1） Compared to paired normal tissue,the expressions of VCP mRNA and PCNA mRNA significantly increased in gastric cancer and lymphatic invasive cancer（P0.01）.Besides,there was correlation between high expression of VCP mRNA and PCNA mRNA and histological differentiation,deep tumor invasion,lymph node metastasis as well as pTNM staging（P0.05）.（2） There was a significant positive correlation between expression of VCP mRNA and PCNA mRNA in gastric carcinoma（r=0.631,P0.01）.Conclusion The high expression of VCP mRNA might be a cause upregulating the PCNA mRNA,and play an important role in tumorigenesis and progression.It might help gastric cancer in predicting prognosis and directing reasonable therapy to detecte their expressions at the same time.
