摘  要:目的建立测定人体内胺碘酮血药浓度的方法。方法以地西泮为内标,用乙酸乙酯提取500μl血样,以KromasilC18柱(5mm×250mm,4.6μm)为固定相,甲醇:水:三乙胺:冰醋酸=80:20:0.1:0.08(体积比)为流动相,流速0.8ml/min,检测波长为240nm,柱温为30℃。结果本方法在0.256~5.12mg/L之间线性良好,回归方程为Y=3.8456X-0.2507,r=0.9951,日内及日间RSD<10%,最低检测浓度为0.128mg/L。结论该方法简便、准确,适用于胺碘酮的血药浓度监测和药动学分析。Objective To develop a method for amiodarone in serum by high performance liuquid chromatography(HPLC).Methods Diazepam was used as the inter-standard,500μl serum sample was extracted by Ethyl acetate,dissolved by mobile phase.Kromasil C18(5mm×250mm,4.6μm)column was used;mobile phase:methanol-water-Triethylamin-aceticacid(80:20:0.1:0.08),flow rate 0.8ml/min.Ultraviolet detection wavelength was 240nm,T=30℃.Results The ratio of the peak area of amiodarone and diazepam against the concentration of amiodarone was linear with a correlation coefficicent of 0.9951 over the range of 0.256~5.12mg/L,Y=3.8456X-0.2507.The RSD of intra-day and the inter-day were less than 10%.The minimal detectatle concentration was 0.128mg/L.Conclusion The method is simple,accurate and precise.It′s usable for clinical pharmacokinetic studies and monitorring of amiodarone in serum.
