摘  要:文章通过微波辅助萃取(MAE),高效液相毛细管电泳法(HPEC)分析黄连生物碱,得到最佳提取条件是:浸泡时间(60min),萃取溶剂(1g样品,5mL),萃取溶剂组成(0.2%盐酸/甲醇),微波功率(400W)和提取时间(5min)。优化后的条停在定量分析中具有很好的精密度(RSD值小于7.50/0)、回收率(从95.4%到112.7%),以及良好线性关系,范围在5.0~500μg/mL为小檗碱(R^2〉0.999)、1.0~100μg/mL为黄连碱(R^2〉0.996)、0.5—50μg/mL为药根碱(R^2〉0.994)。实验表明MAE-HPEC—DAD是一个筒单,快速,低成本的提取分析方法,可以定量分析黄连中主要生物碱。In the work, Microwave-assisted Extraction(MAE) followed by high performance capillary electrophoresis(HPCE) was developed for quantitative analysis of alkaloids in coptidis rhizoma. After systematical investigation, the optimal experimental parameters of soak time (60 min), extraction solvent volume (1 g sample, 5 mL), extract solvent composition (0.2 % HC1/methanol), microwave power (400 W) and extraction time (5 min) were investigated. The optimized method provided satisfactory precision (RSD values less than 7.5 %), good recovery (from 94.4 % to 123 %), and good linear relation in the range of 5.0-500 μg/mL for berberine (R^2〉0.999), 1.0-100 μg/mL for coptisine (R^2〉0.996) and 0.5-50μg/mL for Jatrorrhizine. The proposed MAE HPEC was a simple, rapid and low-cost method for quantitative analysis of alkaloids in Coptidis Rhizoma, and a potential tool for quality assessment of coptidis rhizoma.
