摘  要:目的探讨新生鼠缺氧缺血损伤脑组织水通道蛋白4的蛋白和mRNA(aquaporin-4,AQP-4)表达的变化及意义。方法健康7d龄SD大鼠共80只,分为对照组和缺氧缺血性脑损伤模型组(HIBD组)。每组动物8只。HIBD组经右侧颈总动脉结扎后,吸入8%O21h,建立HIBD模型。对照组仅行假手术,不予颈总动脉结扎和缺氧。HIBD组于HIBD模型制成后0、6、24、48h和72h分别处死动物(各时间点动物8只),对照组与HIBD组相对应时间点分别处死动物,对两组动物进行脑组织病理学观察,Western Blot免疫印迹法检测脑组织AQP-4蛋白的表达和RealtimePCR检测脑组织AQP-4mRNA的表达。结果 HIBD组在HIBD后6、24、48h和72h脑组织病理学出现脑水肿及软化、梗死等改变,并随缺氧缺血后时间延长病理学改变加重。与对照组比较,HIBD组脑组织AQP-4蛋白和AQP-4mRNA表达从HIBD后6h即开始下降,48h内下降至最低,72h出现恢复,但仍低于对照组(P<0.05)。结论 AQP-4可能参与脑内渗透平衡的重建,AQP-4表达下降可能与HIBD脑组织脑水肿及病理学变化的发生发展有关。Objective To explore the expression of AQP-4 protein and AQP-4mRNA in the brain tissue of newborn rat with HIBD.Methods 7-day old SD rats were subjected to the ligation of right carotid artery(ischemia),then they were put into a box full with 8% oxygen and 92% nitrogen for an hour(hypoxia).The SD rats with HIBD model were divided into 5 groups(Each group was 8 rats),including 6、24、48h and 72h after establishment of HIBD.The normal control groups were also divided into 5 groups same as HIBD groups.The Western Blot was used to detect the AQP-4 protein expression and RealtimePCR was used to detect the AQP-4 mRNA expression in the brain tissue of both HIBD and the control groups.Results The brain edema and infarction was present pathologically in brain tissues in HIBD groups after 6h of HIBD and become more severe further with time compared with that in the control groups(P0.05).The expressions of AQP-4protein and mRNA were significantly decreased in HIBD groups during the period of 6~72h of HIBD compared with the control groups in the SD neonatal brain tissues(P0.05).Conclusion It is likely that the decreased expression of AQP-4 is associated with brain edema and brain damage in HIBD and AQP-4 may play an important role in such brain damage conditions.
