RP-HPLC测定川明参药材中香豆素含量

摘　　要：目的建立测定川明参药材中2种香豆素成分欧前胡素及5-异戊烯基-8-甲氧基补骨脂素含量的RP-HPLC测定方法。方法色谱柱:Kromasil-C18(250mm×4.6mm,5μm),流动相为甲醇-水(65∶35),流速为1ml/min,柱温为30℃,检测波长为246nm.结果欧前胡素在0.16～0.97μg(r=0.9998),5-异戊烯基-8-甲氧基补骨脂素在0.13～0.76μg(r=0.9996)范围内与色谱峰面积线性关系良好,欧前胡素和5-异戊烯基-8-甲氧基补骨脂素的平均回收率分别为98.79%、98.11%,RSD分别为1.65%、1.73%.结论该测定方法准确、简便、重复性好。研究结果为建立完善川明参药材质量标准奠定了基础。Objective To establish the determination method of imperatorin and 5-isopentenyl-8-methoxy-psoralen in Chuanminshen violaceum by RP-HPLC.Methods Chromatogram column：Kromasil-C18（250 mm×4.6 mm,5 μm）. The mobile phase was menthol-water （65∶35）; The flow rate was 1.0 ml/min; The column temperature was 30℃; The UV detection wavelength was set at 246 nm.Results There were linear relationship between the absorption area value and the concentration in the range of 0.16～0.97 μg （r=0.9998） for imperatorin and in the range of 0.13～0.76 μg （r=0.9996） for 5-isopentenyl-8-methoxy-psoralen respectively. The average recovery rates were 98.79%（RSD=1.65%） and 98.11% （RSD=1.73%）,respectively.Conclusion The method is accurate,simple and of good reappearance. The research results have laid experimental foundations for the quality control of Chuanminshen violaceum.
