4个鸡品种VIPR-1基因遗传多样性分析

摘　　要：采用PCR-RFLP方法,对茶花鸡、泰和丝羽乌骨鸡、固始鸡和隐性白洛克鸡VIPR-1基因的20个位点的遗传多样性进行研究。结果表明,20个位点的基因频率在这4个群体中分布差异很大,绝大部分的SNP最小等位基因频率均大于5%。独立χ2分析显示,除位点C1704887T外,其他19个位点基因型分布在不同鸡品种间存在显著或极显著差异(P<0.05、P<0.01)。茶花鸡、泰和丝羽乌骨鸡、固始鸡与隐性白洛克鸡4个群体的平均多样性分别为0.3658(±0.0238)、0.3609(±0.0315)、0.3836(±0.0299)和0.3598(±0.0401),显著性检验差异均不显著。单倍型分析发现,在所研究的区域,泰和丝羽乌骨鸡有2个单倍型块,固始鸡有4个单倍型块,茶花鸡和隐性白洛克鸡有5个单倍型块。研究发现鸡VIPR-1基因中5个位点,单倍型块C1660256T～A1660309G以及G1680779T～C1693306T的12kb区域可能与早期生长速度有关。Twenty variation sites in the chicken VIPR-1 gene were genotyped using PCR-RFLP method to study ggenetic diversities of four chicken breeds including Chahua chickens（CH）,Taihe Silkies（TS）,Gushi chickens（GS） and White Recessive Rock Broilers（WRR）.The results indicated that the allele frequencies of 20 sites were significant different among the four chicken populations,moreover,minor allele frequencies of most of SNP were higher than 5%.The chi-square test revealed that there were significant differences of allele frequencies among four chicken populations（P 0.05,P 0.01） for all sites except for C1704887T.The mean diversity of CH,TS,GS,and WRR were 0.3658（±0.0238）,0.3609（±0.0315）,0.3836（±0.0299）,and 0.3598（±0.0401）,respectively.However,there was no significant difference between any two breeds.Based on the four-gametal ruler,the analyzed region of CH,TS,GS,and WRR could be divided into 5 disruptive haploblocks,2 disruptive haploblocks,4 disruptive haploblocks,and 5 disruptive haploblocks,respectively.Generally,2 to 3 haplotypes within each block could represent for over 90% diversity information.This study predicted that five SNP,haplotype block based C1660256T and A1660309G,12 kb region from G1680779T to C1693306T might be associated with early rate of growth.
