裸鼠人胃癌MKN-45细胞核转录因子NF-κBP^(65)蛋白与增殖细胞核抗原表达的相关性

摘　　要：目的:通过观察胃癌细胞核转录因子NF-κBP65蛋白与增殖细胞核抗原(PCNA)蛋白的表达及关系,探讨消痰散结方抑制胃癌细胞增殖的作用机制。方法:采用OB胶粘贴法建立裸鼠人胃癌MKN-45原位移植瘤模型,随机数字表法分成模型组,消痰散结低、中、高剂量组,5-Fu组,每组8只,观察消痰散结方干预对胃癌组织核转录因子NF-κBP65蛋白、增殖细胞核抗原(PCNA)表达的影响及相关性。结果:①消痰散结方中、高剂量组、5-Fu组NF-κB的P65蛋白的阳性表达与模型组相比P<0.05,有统计学意义;②消痰散结方低、中、高剂量组、5-Fu组的PCNA阳性表达率与模型组相比有统计学意义,P<0.05;③胃癌细胞NF-κBP65蛋白与PCNA表达存在正相关(r=0.857,P<0.01)。结论:胃癌MKN-45细胞中存在着活化的NF-κB及高表达的PCNA,并且存在线性相关,消痰散结方可能通过其抑制NF-κB活性,阻断了NF-κB对PCNA的活性调控,使胃癌细胞增殖受到抑制,控制胃癌复发与转移。Objective： To discover the mechanism of Xiaotan Sanjie Fang （XTF） , a traditional Chinese medicinal empirical Prescription for curing gastric cancer, in inhibiting proliferation of gastric cancer cells by obverving the expression and relation between gastric Cancer cell NF - κBP^65 and PCNA protein. Methods： Using OB glue paste method, the nude mice human gastric cancer MKN -45in situ transplantation tumor model was established, based on which 40 animals were randomly divided into 5 groups（ 8 mice/every group） ： the blank model group, low -dose XTF group, intermediate -dose XTF group, high - dose XTF group, and 5 - Fu group, to observe the influence and relation of Xiaotan Sanjie Fang intervening gastric cancer cell NF - κBP^65 and PCNA protein. Results ：（1）There was a statistical meaning by comparing the intermediate -, high - dose XTF groups and 5 - Fu group on NF - κBP^65 protein positive expression rate with the model group（ P 〈0.05）. （2）There was a statistical meaning by comparing the low -, intermediate - , high -dose XTF group and 5 -Fu group with the model group on PCNA protein positive expression rate（ P 〈 0.05 ）. （3）NF - κBP^65protein and PCNA protein expression in gastric Cancer cells is a positive correlation （ r = 0. 857, P 〈 0. 01 ）. Conclusion ： There are activated NF - κBP^65 protein and PCNA protein in the gastric cancer MKN - 45 cell, and show linear correlation. Xiaotan Sanjie Fang can betterly inhibit gastric cancer cells proliferation and control of gastrol cancer recurrence and metastasisby restraining NF- κBP^65 protein activity, and blocking NF- κBP^65 protein to regulate PCNA protein activity.
