摘 要:目的:以益肾活血清利为法组方通过中药含药血清观察中药对HK-2细胞(人肾近端小管上皮细胞)TEMT(肾小管上皮细胞向间质肌成纤维细胞转分化)过程中的信号因子表达的影响。方法:制备中药含药血清,观察含药血清对HK-2细胞在TGF-β1诱导下EMT过程的阻抑作用。随机分为正常组、模型组、中药低剂量组、中药高剂量组、苯那普利组。观察在TGF-β1诱导下转分化过程中对Smad4及RhoA mRNA表达作用的影响。结果:中药低剂量组、苯那普利组与模型组Smad4的mRNA表达比较有显著差异;中药高剂量组RhoA的mRNA表达与模型组比较P<0.05,差异有显著性;而中药低剂量组、苯那普利组与模型组比较差异不显著。结论:在Smad4 mRNA的表达上中药低剂量组与洛汀新均显示出抑制作用,但洛汀新对RhoA mRNA的表达上无明显作用,而中药高剂量组则有明显的抑制RhoA mRNA表达的作用,说明中药的可探讨性较大,单纯通过剂量调整或许可以达到多信号途径的抑制作用而发挥拮抗EMT的过程。Obiective:To identify whether Yishen Huoxue Qingli recipe plays an important role in vitro in preventing the development of EMT in HK2 cells.Methods:The cells were randomly divided into five groups:normal group,model group,the yishen huoxue qingli Chinese medicine group,the high dosage Chinese medicine group and the benazepril group.HK2 cells were induced by TGF-β1,Through the blood serum with traditional Chinese medicine,we observe the expression of smad4 and RhoA mRNA in progress of the EMT in HK2 cell stem.Results: The Chinese medicine group and the benazepril group significantly inhibited the expression level of Smad mRNA(P〈0.05),the high dose Chinese medicine group was not significant;The high dose Chinese medicine group significantly inhibited the expression level of RhoA mRNA(P〈0.05),while the Chinese medicine group and the benazepril group were not significant.Conclusion:we noted that the expression of smad4 and RhoA mRNA in the Chinese medicine group and the high dose Chinese medicine group is better than that in the benazepril group.The benazepril group only inhibites smad4 mRNA.
