硫酸软骨素酶ABC对大鼠脊髓损伤后生长相关蛋白43和神经胶质纤维酸性蛋白的影响  被引量:7

摘  要:目的探讨硫酸软骨素酶ABC(chondroitinase ABC,ChABC)对大鼠脊髓损伤(spinal cord injury,SCI)后生长相关蛋白43(growth associated protein43,GAP-43)和神经胶质纤维酸性蛋白(glial fi brillary acidic protein,GFAP)表达的影响。方法取成年雌性SD大鼠150只,体重250~300g,随机分为ChABC治疗组(A组)、生理盐水治疗组(B组)和假手术组(C组),每组50只。A、B组制作大鼠脊髓横断损伤模型,分别于SCI区通过蛛网膜下腔注射6μL ChABC(1U/mL)和等量生理盐水治疗;C组不切断脊髓及不治疗作为空白对照。于术后1、3、7、14、21d采用BBB评分法观察大鼠后肢运动功能情况,用免疫荧光染色法检测各组GAP-43和GFAP阳性表达并计数阳性表达神经元数量。结果术后各时间点A、B组BBB评分均明显小于C组(P<0.05)。术后1、3、7d,A、B组间BBB评分差异无统计学意义(P>0.05);术后14、21d,A组BBB评分明显高于B组(P<0.01)。免疫荧光染色示,术后各时间点A、B组GAP-43和GAFP阳性神经元数均明显多于C组(P<0.05)。术后14、21d,A组GAP-43阳性神经元数明显多于B组(P<0.01);1、3、7d时两组比较差异无统计学意义(P>0.05)。术后7、14、21d,A组GFAP阳性神经元数明显少于B组(P<0.01);术后1、3d时两组比较差异无统计学意义(P>0.05)。结论 ChABC能够降解胶质瘢痕,改善大鼠SCI区域的微环境,使GAP-43表达增强,促进神经轴突的生长与延伸。Objective To investigate the effect of chondroitinase ABC (ChABC) on the expression of growth associated protein 43 (GAP-43) and glial fibrillary acidic protein (GFAP) after spinal cord injury (SCI) in rats. Methods A total of 150 adult female SD rats, weighing 250-300 g, were randomly divided into ChABC treatment group (group A), saline treatment group (group B), and sham operation group (group C) with 50 rats in each group. In groups A and B, the rats were made the SCI models and were treated by subarachnoid injection of ChABC and saline; in group C, the rats were not treated as a control. At 1, 3, 7, 14, and 21 days after operation, the Basso, Beattie, and Bresnahan (BBB) score system was used to evaluate the motion function, and immunofluorescent histochemical staining was used to observe the expressions of GAP-43 and GFAP. Results At different time points, the BBB scores of groups A and B were significantly lower than those of group C (P 〈 0.05); there was no significant difference in BBB score between groups A and B after 1, 3, and 7 days of operation (P 〉 0.05), but the BBB score of group A was significantly higher than that of group B after 14 and 21 days of operation (P 〈 0.01). At different time points, the GAP-43 and GFAP positive neurons of groups A and B were significantly higher than those of group C (P 〈 0.05). After 14 and 21 days of operation, the GAP-43 positive neurons of group A were more than those of group B (P 〈 0.01 ). After 7, 14, and 21 days of operation, the GFAP positive neurons of group A were significantly less than those of group B (P 〈 0.01). Conclusion ChABC can degrade glial scar, improve the microenvironment of the injured region and enhance the expression of GAP-43, which promotes axonal growth and extension.
