摘  要:目的探讨高压氧(HBO)治疗对继发性脊髓损伤(SCI)的保护作用及其机制。方法96只SD大鼠采用Allen打击法造成SCI后分为HBO组、常氧高压氮组、常压吸氧组和常压空气组,分别于损伤后第1,3,7,14天取材,采用原位末端标记法(TUNEL)检测凋亡细胞,PV-9000二步法免疫组化检测caspase-3表达,光镜下观察,并对结果进行统计学分析;神经功能评价采用BBB评分。结果HBO组在各时间点easpase-3表达均低于其他各组;TUNEL结果亦显示,HBO组凋亡细胞数低于其他各组;HBO组大鼠神经功能明显改善。结论HBO能通过下调caspase-3的表达抑制SCI后细胞凋亡,其作用与氧有关。Objective To investigate the mechanism underlying the protective effect of hyperbaric oxygen (HBO) in secondary spinal cord injury (SCI). Methods Models of acute SCI were established in 96 Sprague- Dawley rats using Allen's dropping weight technique. The rats were then divided into a HBO group, a l^igh pressure nitrogen normal oxygen group, a normal pressure oxygen group and a normal pressure air group. The injured spinal cords were sampled for morphological studies at the 1 st, 3rd, 7th and 14th day after injury. Apoptotic cells were la- beled using the TUNEL technique, and the expression of caspase-3 was detected. The neurological functionality of the spinal cord was assessed by open field locomotor evaluation (the BBB score). Results The expression of caspase- 3 in the HBO group decreased significantly more than in the other groups after injury. The number of TUNEL-positive cells was significantly lower in the HBO group as well. Neurological function improved significantly after HBO thera- py. Conclusions HBO can down-regulate the expression of caspase-3 and inhibit cell apoptosis in rats after SCI. The protective effect of HBO was related with the oxwen level.
