摘  要:目的 观察诱导型一氧化氮合酶(iNOS)在川芎嗪(TMP)拮抗顺铂(CDDP)耳毒性中的表达情况,探讨顺铂耳毒性的致病机制及川芎嗪对顺铂耳毒性的防护作用.方法 选取50只健康豚鼠,随机分为对照组、顺铂组和中药组,各组动物均于用药前及停药后测试听觉脑干诱发电位(ABR),利用免疫组织化学方法测定各组耳蜗内iNOS的表达情况.结果 对照组ABR阈值(40.94±6.75)db,顺铂组ABR阈值为(76.26±4.54)db,中药组ABR阈值为(58.98±5.82)db,3组比较差异具有显著性(P〈0.01);免疫组化结果显示iNOS在对照组不表达,顺铂组Corti氏器和螺旋神经节细胞胞浆内有强阳性表达,中药组有阳性表达,3组比较差异具有显著性(P〈0.01).结论 川芎嗪可以通过抑制iNOS的表达来对抗顺铂的耳毒性.Objective To observe the expression of iNOS in the experiment of ligustrazine against cisplatin-induced,and to investigate pathogenic mechanism and protection of ligustrazine against eisplatin-indueed ototoxicity. Methods Fifty white guinea pigs with normal hearing were divided into three groups randomly, that is control group, experimental group and an inhibited group. The alternations of hearing function were evaluated by the examination of auditory brainstem response ( ABR ) and the latency of wave. The immunohistoehemistry was used to detect the expression of induced nitricoxide synthase( iNOS ) in the specimens. Results Compared with control group,the threshold of ABR in experimental group and inhibited group were significantly different. There was a significant difference between the experimental group and the inhibited group. There was no expression of iNOS in Cortis, spiral ganglion cell and stria vascularis in normal cochlear. In the experimental group, the expression of iNOS became stronger in spiral ganglion cell, stria vascularis, spiral ligament and Cortis: Ligustrazine reduced the expression of iNOS induced by cisplatin. Conclusion Ligustrazine could be used to treat the cisplatin-induced ototoxieity and prevent the chemotherapy complication.
