摘  要:为了获得本地毛形线虫(Trichinella nativa)49 ku ES蛋白的结构基因,用Trizol提取T.nativa肌幼虫的总RNA,用RT-PCR方法扩增出了编码T.nativa49 ku排泄分泌蛋白的结构基因。将PCR产物与T载体连接并经过大肠埃希菌增殖后送检测序。序列测定表明,目的基因TNPG长度为951bp,核苷酸序列同已发表的T.spiralis相应的序列P49同源性为98.63%,所推导的氨基酸序列同源性为99.05%。为T.nativa49 ku ES蛋白结构基因的进一步研究打下基础。The target gene of 49 ku ES protein of T.nativa was searched here.The total RNA of Trichinella nativa was isolated using Trizol after the worms being pestled.The gene of the worm which encoding 49 ku ES protein was amplified by RT-PCR.PCR products were sequenced after being linked with T-vector and propagated by E.coli.It was shown from the sequence analysis that the target gene TNPG was consisted of 951 bp and 98.63% nucleotide homology to the sequence of P49 from T.spiralis,and 99.05% homology of deduced amino acid between TNPG and P49.This study lays a base for the more research on the 49ku ES protein of T.nativa.
