禽B型白血病病毒的分离及分子分型鉴定

摘　　要：从送检的骨石症蛋用种鸡中分离出一株禽白血病病毒(ALV)。剖检骨石症发病鸡,采集病变的肝脏、脾脏组织,提取病毒基因组,并利用针对ALV群特异性抗原P27基因设计的引物进行PCR快速检测确定为ALV感染。将病变组织接种SPF鸡胚绒毛尿囊膜,连续传代3次。进一步针对亚群间特异性抗原gp85区设计一对引物进行PCR扩增并酶切分析。结果确认分离到的1株病毒为ALV-B亚型。A strain of avian leukosis virus was isolated from the submitted layer chickens with osteopetrosis.First,the chickens was anatomized,and their pathological livers and spleens were collected to extract the genomes of virus.The result of rapid detection of PCR,which primers were designed by P27 gene,showed that the specific antigens of ALV groups,were determined.Then,pathological tissues were disposed carefully and inoculated in the chorioallantoic membrane of chick embryo,and subcultured for three generations.Furthermore,the couples of primers were designed by gp85 gene segment,the specific antigen of subgroup,to conduct the PCR amplification and identification of restriction enzyme digestion.The result showed the subtype of isolated virus was ALV-B.
