摘 要:利用ISSR分子标记技术对蒙古冰草与航道冰草正反交杂种染色体加倍植株F2代间及其与亲本和杂种F1代间在DNA分子水平上的遗传差异性进行分析,结果表明:12个ISSR适宜引物共扩增出515个条带,多态性条带492个,多态性条带百分率为95.3%。6个供试材料间的遗传距离(GD)变幅为0.4094-0.6721,平均为0.5391,亲本航道冰草与蒙古冰草间、航道冰草与正反交杂种染色体加倍植株F2间的遗传距离较大,正反交F1间的遗传距离次之,正反交杂种染色体加倍植株F2间的遗传距离相对较小。以GD值0.51为基准,6个材料聚为3类:第一类为蒙古冰草、反交杂种F1,第二类为正交杂种F1、正反交杂种加倍植株F2,第三类为航道冰草。To determine the degree of genetic difference among chromosome doubling plants F2 of reciprocal hybrids F1 between Agropyron mongolicum and A.cristatum cv.Fairway,their parents and reciprocal hybrids F1,and provide basis for selection of good lines from tetraploid chromosome doubling progenies,the polymorphism among the tested plants were analyzed based on ISSR molecular marker.The results showed that 515 bands of ISSR markers were amplified with 12 suitable primers,492 bands were polymorphic,and the percentage of polymorphism bands reached 95.3%.The genetic distance(GD) of 6 tested plants were ranged from 0.4094 to 0.6721,and the average of genetic distance was 0.5391.The genetic distance between A.cristatum cv.Fairway and A.mongolicum,between A.cristatum cv.Fairway and chromosome doubling plants F2 of reciprocal hybrids was wide,and then the genetic distance between Agropyron mongolicum × A.cristatum cv.Fairway F1 and A.cristatum cv.Fairway ×A.mongolicum F1,and the genetic distance among chromosome doubling plants F2 of reciprocal hybrid was the nearest.Using the GD value of 0.51 as threshold,6 tested plants could be clustered into 3 groups: the first group including A.mongolicum and inverse cross F1,the second groups including obverse cross F1 and chromosome doubling plants F2 of reciprocal hybrids,and the third groups including A.cristatum cv.Fairway.
