摘 要:目的应用脱氧核苷酸转移酶介导的原位末端标记技术(TUNEL),检测咖啡酸锗对小鼠宫颈癌U14瘤的凋亡诱导作用。方法建立小鼠宫颈癌U14移植瘤动物模型,观察抑瘤率,并应用TUNEL检测各组瘤细胞凋亡阳性率。结果咖啡酸锗对小鼠有明显抑制作用,其高、中、低剂量分别为45.47%、57.86%、55.83%,差异有统计学意义(P<0.01),TUNEL显示咖啡酸锗低剂量组细胞凋亡阳性率最高,达80.00%,与对照组比较差异有统计学意义(P<0.01)。结论咖啡酸锗对小鼠U14瘤有明显的抑制作用,可诱导U14移植瘤细胞凋亡,可能是其抑癌机制之一。Objective To use situ terminal deoxylnucleotidyl transferase mediated-UTP nick end labeling(TUNEL)for detecting the apoptosis effect induced by caffeic acid Ge on rat cervical carcinoma U14.Methods To establish the rat model of transplantable cervical carcinoma U14.The tumor inhibition rate was observed and the positive rate of apoptosis in various groups of tumor cells was detected by TUNEL.Results Caffeic acid Ge had the significant inhibitory effect on rats.The positive rate of apoptosis in high,middle and low dose groups were 45.47%,57.86% and 55.83%,respectively(P〈0.01).The TUNEL method showed that the positive rate of apoptosis in low dose group was the lowest,reaching 80.00% with statistical difference cpmpared with the contol group(P〈0.01).Conclusion Caffeic acid Ge has the significant inhibitory effect on rats cervical carcinoma U14 and induces the apoptosis of transplantable tumor U14,which could be the one of suppression tumor mechanisms
