摘  要:从2008年送检的病猪病料中分离到1株致细胞病变代次、时间以及细胞病变形态与典型高致病性猪繁殖与呼吸综合征病毒(HP-PRRSV)培养特性不同的PRRSV(LD812株)。对LD812株的Nsp2和ORF5基因进行测序发现,该株病毒与标准HP-PRRSVJXA1株(与VR2332相比,JXA1Nsp2基因的第478、538~566位氨基酸缺失,缺失形式为1+29个氨基酸)的Nsp2基因同源性为98.3%,LD812分离株Nsp2基因上第475~520和538~566位氨基酸缺失,缺失形式为46+29个氨基酸,比JXA1株在1号缺失位多缺失45个氨基酸;LD812株ORF5基因序列与JXA1株相比仅有8个核苷酸的点突变。结合该毒株的临床特征、细胞培养及分子特性,推测LD812为一弱毒株。An specific porcine reproductive and respiratory syndrome virus(PRRSV) LD812 strain was discovered from swine clinical samples of Hunan province in 2008,which represented different characteristics in pathogenic generations,latent period and pathological changes compared with other previous reports on Chinese PRRSV isolates.The complete sequences of Nsp2 gene possessing cysteine protease activity for autocleavage and the envelope glycoprotein gene(ORF5) were determined.Comparison between LD812 and the highly pathogenic PRRSV standard strain JXA1 showed 98.3% nucleotide identity(1 and 29 aa deletions are conserved in JXA1 isolate in the aa position of 478 and 538-566 compared with the VR2332 strain),the Nsp2 gene of LD812 has a 46 deletions in the aa position of 475-520 and a 29 deletions in 538-566 which suggested a novel 45 deletions in aa position of 478.The ORF5 gene of LD812 nearly has no mutation contrasted with JXA1 isolate.The LD812 is recognized as a low virulent strain according its cultural and clinical characteristics.
