摘  要:探讨中药单体成分β-谷甾醇(β-sitosterol)、淫羊藿苷(Icariin,Ica)对猪精子体外获能的影响,以期提高体外受精效果。以mTBM+肝素获能液为对照组,分别添加β-谷甾醇、淫羊藿苷的低、中、高3个剂量为试验组,利用金霉素荧光染色(Chlortetracycline,CTC)法结合体外受精检验体外获能效果。在mTBM+肝素中添加β-谷甾醇2(中剂量0.08mg/L)、淫羊藿苷2(中剂量17mg/L)的B型精子率(获能精子)显著高于对照组和其他剂量组(P<0.05),F型精子率(未获能精子)显著低于对照组和其他剂量组(P<0.05),AR型精子率(发生顶体反应精子)与对照组和其他剂量组差异不显著(P>0.05),卵裂率和囊胚率均极显著高于对照组和其他剂量组(P<0.01);表明中药单体成分β-谷甾醇、淫羊藿苷能够显著促进猪精子体外获能效果。To investigate the effect of adding two kinds of chinese tradional madicine monomer β-sitosterol and icariin on porcine spermatozoa capacitation in vitro.The discrepancy effect on porcine spermatozoa capacitation in vitro between β-sitosterol and icariin was compared by using chlortetracycline (CTC) staining and in vitro fertilization (IVF).The results showed that β-sitosterol 2(0.08 mg/L) and icariin 2(17 mg/L) groups could improve the effect of capacitation in vitro.In experiment 1,the rate of porcine spermatozoa capacitation in vitro (38.53%) in β-sitosterol 2 group was higher than the other groups.The cleavage rate (76.32%),blastocyst rate (24.50%) of β-sitosterol 2 group were extremely higher than the other groups.There was no statistical difference between the rest (P0.05).In experiment 2,the rate of porcine spermatozoa capacitation in vitro (40.89%) in Ica 2 group was higher than the other groups.The cleavage rate (78.58%),blastocyst rate (26.67%) of Ica 2 group were extremely higher than the other groups.There was no statistical difference between the rest of groups(P0.05).In conclusion,the results showed that adding two kinds of Chinese tradional madicine monomer β-sitosterol and icariin could obviously improve the effect of porcine spermatozoa capacitation in vitro.
