摘 要:应用荧光光谱法和紫外-可见吸收光谱法研究了穿心莲内酯(ANDRO)与牛血清白蛋白(BSA)之间的相互作用。实验表明,ANDRO对BSA的荧光猝灭作用是一种静态猝灭过程,在生理条件下二者约以1∶1相结合形成复合物。在不同温度下得到ANDRO与BSA间结合反应的平衡常数,并与通过紫外-可见吸收光谱法得到的结合常数进行对照,证明结论可靠。根据F rster非辐射能量转移理论,求算了相互作用体系中给体与受体间的距离为4.37 nm,并通过体系热力学参数推出ANDRO与BSA间主要为疏水作用力。最后利用同步荧光光谱探讨了ANDRO对BSA构象的改变,并讨论了几种金属离子对ANDRO-BSA相互作用体系的影响。The interaction between andrographolide(ANDRO) and bovine serum albumin(BSA) was studied by fluorescence and ultra-violet absorption spectroscopic method.The results indicated that the reaction of ANDRO with BSA was a static quenching process and there was a ground-state complex that was formed with the molar ratio of 1∶1 under the physiological conditions.At different temperatures,the apparent association constants of the reaction between ANDRO and BSA were obtained,and the reliability of the data was verified by UV-vis spectrometry.The distance between acceptor and donor was calculated on the basis of Frster theory and,that was 4.37 nm.The force between ANDRO and BSA is an electrostatic force determined by analyzing thermodynamic parameters.Then the effect of ANDRO on the conformational change of BSA was investigated by synchronous fluorescence spectroscopic method,and the influences of the presence of some metal ions on ANDRO-BSA system were discussed.
